摘要 目的:探究牡丹叶治疗原发性痛经的多成分、多靶点、多途径作用机制,尝试建立其可能的"药物-核心成分-关键靶标-主要通路"的网络药理研究模式。方法:针对牡丹叶中15个入血成分,利用中药整合药理学计算平台建立牡丹叶调节疾病的候选靶标网络图,再根据网络拓扑特征参数值筛选出牡丹叶治疗疾病的关键靶标名称及其所参与的通路,并最终绘制出"药物-核心成分-关键靶标-主要通路"多维网络关系图。结果:预测实验表明,15个入血成分的作用靶点约为299个,共涉及主要通路20条与原发性痛经相关。结论:牡丹叶可能通过作用于镇痛、解痉、抗炎、激素调节以及免疫调节等相关的蛋白靶点和通路起到治疗原发性痛经的作用。 Objective: To explore the multi-component,multi-target and multi-pathway mechanism of peony leaf in the treatment of primary dysmenorrhea,and try to establish the possible network pharmacological research model of " drug-core component-key target-main pathway". Methods: In view of the 15 blood-entry components in the leaves of peony,the network diagram of the candidate target of peony solution was established by using the integrated pharmacology platform of Chinese medicine. The key target name of peony leaf and all the pathways involved were selected according to the parameters of the network topology. Finally, " multidimensional network relation diagram" of " drug-core component-key target-main pathway" was drawn. Results: Prediction experiments showed that there were about 299 targets among the 15 blood-entry components,involving 20 main pathways related to primary dysmenorrhea. Conclusion: Peony leaves may play a role in the treatment of primary dysmenorrhea by acting on analgesic,spasmodic,anti-inflammatory,hormonal regulation,and immunoregulation.
