基于生物信息学的条斑紫菜microRNAs及其靶标预测 被引量:1

摘要 目前,作为模式红藻的条斑紫菜面临着日益严峻的生存危机。非编码RNA介导的基因后转录水平调控对于条斑紫菜资源的保护与开发具有十分重要的意义。研究利用BLASTn结合Mireap软件,共预测到5条新的条斑紫菜miRNAs;利用Target Finder工具预测到5条与紫菜生长发育及代谢相关的靶标基因,另预测到59条未知功能的靶标位点。Clustal W结合WebLoGO的分析结果显示了条斑紫菜miRNAs在植物间保守性;Clustal W结合MEGA的miR164家族系统进化结果显示,pre-miR64s与物种进化不同步,且具有进化的随机性与序列的多态性。研究结果丰富了条斑紫菜miRNAs及其靶标信息,可为条斑紫菜后转录水平的分子选育及其功能的开发与利用奠定基础。 At present, the stripe laver, which is a model of red algae, faces an increasingly severe crisis of survival. Non-coding RNA-mediated post-transcriptional regulation of genes is of great significance for the protection and development of Porphyra yezoensis resources. In this study, BLASTn and Mireap software were used to predict five new miRNAs of Porphyra yezoensis. Target Finder was used to predict five target genes related to the growth, development and metabolism of Porphyra, and 59 target sites with unknown functions were predicted. The analysis of Clustal W combined with WebLoGO showed that the miRNAs of Porphyra yezoensis were conserved among plants. The phylogenetic analysis of miR164 family based on Clustal W and MEGA showed that the pre-miR164 s were not synchronized with species evolution and had evolutionary randomness and sequence polymorphism. The results enriched the miRNAs and their targets of Porphyra yezoensis, which laid the foundation for the molecular breeding of post-transcription levels of Porphyra yezoensis and the development and utilization of its functions.
