摘要
以野生金银花当年生茎段为外植体,MS和1/2MS作基本培养基,研究不同激素对金银花外植体灭菌消毒效果、不定芽诱导、增殖培养和组培苗生根诱导的影响,以建立野生金银花高效快繁体系。结果表明:外植体最佳消毒灭菌方法为75%碱化乙醇+0.1%升汞消毒12min,外植体无菌苗率达到82.67%;不定芽诱导最佳培养基配方为MS+6-BA 1.5mg/L+NAA 0.12mg/L,不定芽诱导率为88.67%;继代增殖最佳培养基配方为MS+6-BA1.5mg/L+NAA 0.02mg/L,增殖倍数达到4.11,与其他处理差异达到极显著水平(P<0.01);生根诱导培养基配方以1/2MS+IBA 2.5mg/L+NAA 0.2-0.6mg/L较好,生根率和根长分别达到85.56%-92.22%和3.14-3.27cm。
With stem segments of wild Lonicera japonica Thunb.(honeysuckle) as explants,MS and 1/2MS as the basic medium,the effects of different hormones on honeysuckle in terms of explant sterilization effect,adventitious bud induction,proliferation and rooting induced effects were examined,in order to establish a high efficient propagation system of wild honeysuckle.The results show that the best explant sterilization method was 75% ethanol +0.1% alkalization mercuric chloride for 12min,explant aseptic seedling rate reached 82.67%; the best adventitious bud induction medium was MS+6-BA 1.5mg/L+NAA 0.12mg/L,the induction rate of adventitious buds was 88.67%; the best subculture medium was MS+6-BA 1.5mg/L+NAA 0.02mg/L,the proliferation ratio reached 4.11,presenting a significant difference compared to other treatments( P 〈0.01); rooting medium with 1/2MS+ IBA 2.5mg/L+NAA 0.2-0.6mg/L presented well,with the rooting rate and root length 85.56%-92.22% and 3.14-3.27cm,respectively.
