复元醒脑汤对糖尿病脑梗死大鼠神经缺损行为及糖脂代谢的干预作用 被引量:1

摘要 目的:观察复元醒脑汤对糖尿病脑梗死大鼠神经缺损行为以及糖脂代谢的干预作用。方法:将健康雄性SD大鼠150只按随机数字表法分为正常组、假手术组、模型组及西药、中药治疗组,每组30只。腹腔注射链脲佐菌素(STZ)50 mg/kg后予高能量高脂肪饲料喂养2周以复制糖尿病大鼠模型,再采用线栓法制备糖尿病脑梗死模型,造模后中药治疗组灌胃复元醒脑汤10.4 g/kg(体质量),2次/d,连续14天。西药治疗组灌胃二甲双胍27 mg/kg(体质量),2次/d;尼莫地平2.16 mg/kg(体质量),2次/d,连续14天。模型组、假手术组予等量生理盐水灌胃,连续14天。14天后处死大鼠取材,测定大鼠血糖、血清胆固醇、甘油三酯浓度,计算神经缺损行为体征(Bederson)评分。结果:1)与模型组相比,中药治疗组大鼠胆固醇、甘油三酯和血糖水平都明显下调(P<0.01);西药治疗组大鼠血糖水平明显下调(P<0.01);2)中药治疗组和西药治疗组大鼠Bederson评分较模型组显著降低(P<0.01)。结论:复元醒脑汤可降低糖尿病脑梗死大鼠血糖、血脂水平,改善神经缺损行为体征评分。 Objective: To observe the intervention effects of FuYuan XingNao decoction on neurological function and glycolipid metabolism of the rats with diabetic cerebral infarction (CI). Methods: All 150 SD rats were separated into the normal group, sham operation group, the model group and the trealment groups of western medicine and herbs according to random number table method, 30 rats each group. After peritoneal injection of streptozotocin (STZ), 50mg/kg, diabetic rat models were duplicated by feeding them high energy high fat fodder for two weeks, CI models were established by using suture method, after modeling, the treatment group of herbs received intragastric administration of FuYuan XingNao decoction, 10.4g/kg (body mass), twice each day, for 14 days. The treatment group of western medicine were lavaged with melbine (DMBG), 27 mg/kg(body mass), twice each day; nimodipine, 2.16mg/kg (body mass), twice each day, for 14 days. The model group and sham operation group accepted intragastric administration of the same amount of physiological saline for 14 days. The rats were sacrificed in 14 days, the concentrations of blood glucose, serum cholesterol (SC), glycerin trilanrate (GT) of the rats were detected, Bederson scales were calculated. Results: 1) Comparing with the model group, the levels of SC, GT and blood glucose of the rats in the treatment group of herbs down regulated obviously (P〈0.01); the levels of blood glucose of the rats in the treatment group of western medicine lowered notably (P〈0.01); 2) Bederson scales of the rats in the treatment groups of herbs and western medicine were lower than these of the model group (P〈0.01). Conclusion: FuYuan XingNao decoction could decrease the levels of blood glucose and blood lipid of the rats, and improve the scores of neural deficit behavior signs.
