摘要
目的:建立砂仁总酚含量的快速测定方法,比较不同产地砂仁总酚的抗氧化活性,为砂仁药材资源的综合开发和品质评价提供理论基础。方法:以香草酸为对照品,采用福林酚比色法测定砂仁的总酚含量;通过DPPH法、ABTS法和FRAP法测定不同产地砂仁的抗氧化活性。结果:砂仁总酚含量测定的线性范围为0.002 0~0.011 9 mg·mL^(-1),平均加样回收率为96.6%(RSD=3.3%,n=6);3种方法测定的砂仁抗氧化活性均与总酚含量呈显著正相关(P<0.01),相关系数分别为0.966,0.966和0.927。结论:本文建立的砂仁总酚含量测定方法简便、快速、重复性好;不同产地的砂仁均具有一定的抗氧化活性,总酚含量与产地具有一定的相关性。
Objective: To establish a method for rapid determination the content of total phenol and determine the antioxidant activities of total phenol in Fructus Amomi from various habitats. It aims providing theoretical reference for the development and utilization and comprehensive evaluation of Fructus Amomi. Methods: Folin-Ciocalteu colorimetry was adopted to determine the content of total phenol in Fructus Amomi by taking vanilinic acid as a reference. DPPH assay,ABTS assay and FRAP assay were employed to analyze the antioxidant activities. Results: The concentration of vanillic acid had a good linear relationship in range of 0. 002 0-0. 011 9 mg·mL^(-1). The average recovery was 96. 6%( RSD = 3. 3%,n = 6). The antioxidant activities determined by three different methods were all significantly and positively correlated with total phenol( P 〈0. 01).The correlation coefficients were 0. 966,0. 966 and 0. 927,respectively. Conclusion: The established method for determination of total phenol of Fructus Amomi was simple,rapid and with good repeatability. The Fructus Amomi from various habitats had antioxidant activity certainly. And there is some correlation between total phenolic content and habitats.
