胃癌组织中缺氧诱导因子-1α和葡萄糖转运蛋白-1和下游转录因子1的表达及意义 被引量:3

Expression and significance of HIF-lα,GLUT-1 and Gli1 in gastric carcinoma tissues

摘要 目的探讨胃癌组织中缺氧诱导因子-1α(HIF-lα)、葡萄糖转运蛋白-1(GLUT-1)和下游转录因子1(Gli1)的表达及其与临床病理特征的关系。方法选取2015年6月至2017年3月间武汉大学人民医院汉川医院收治的60例胃癌患者,手术切除取胃癌组织为实验组,取距癌缘>1cm的正常胃组织为对照组,制备石蜡切片。采用免疫组化SP法测定两组标本中的HIF-lα、GLUT-1和Gli1的表达情况,比较表达差异,分析3种蛋白表达与患者临床病理特征的关系及对胃癌早期诊断的评估作用。结果实验组胃癌组织标本中HIF-lα、GLUT-1和Gli1的阳性表达率均高于对照组,差异均有统计学意义(均P<0.05)。实验组胃癌标本中HIF-lα、GLUT-1和Gli1的阳性表达率与患者的p TNM分期、分化程度及淋巴结是否转移有关,差异均有统计学意义(均P<0.05),与性别、年龄及肿瘤直径无关,差异均无统计学意义(P>0.05)。3种蛋白两两之间均存在正相关关系,差异均有统计学意义(P<0.05);受试者工作特征(ROC)曲线分析显示,3种蛋白的ROC曲线下面积(AUC)均在0.6附近或更高。结论胃癌组织中HIF-lα、GLUT-1和Gli1均呈高表达,且三者呈正相关关系,可共同促进胃癌的发生发展,或可作为胃癌早期诊断及预后效果评估的重要参考指标。 Objective To study the expression of hypoxia-inducible factor-1α( HIF-lα),glucose transporte-1( GLUT-1) and downstream transcription factor( Gli1) in gastric carcinoma tissues and the relationship with clinicopathologic features. Methods A total of 60 patients with gastric cancer treated at Wuhan University People's Hospital from June 2015 to March 2017 were selected as research subjects. The normal gastric tissues 1 cm apart from the margin of cancer were chosen as the control group and then paraffin sections were prepared. The expression of HIF-lα,GLUT-1 and Gli1 in the two groups were measured by immunohistochemical SP method. Differences in the expression of the three proteins between the two groups were compared. The relationship between the expression of three proteins and clinicopathologic features were analyzed. Its significance in the early diagnosis of gastric cancer was evaluated. Results The positive expression rates of HIF-lα,GLUT-1 and Gli1 in gastric cancer tissue specimens were significantly higher in the experimental group than in the control group( all P〈0. 05). The positive expression rates of HIF-lα,GLUT-1 and Gli1 of gastric cancer tissue specimens in the experimental group were related with p TNM staging,differentiation degree and lymph node metastasis( all P〈0. 05) rather than with gender,age and tumor diameter( all P〈0. 05). There was a positive correlation between each other among the three proteins( all P〈0. 05).ROC analysis showed that the area of AUC under the ROC curve for the three proteins was near 0. 6 or higher.Conclusion HIF-lα,GLUT-1 and Gli1 are highly expressed in gastric cancer tissues. There is a positive correlation among the three proteins,which may promote the occurrence and development of gastric cancer,and can be used as an important reference index for early diagnosis and prognosis evaluation of gastric cancer.
