地稔预处理对大鼠肢体缺血再灌注损伤的保护作用 被引量:7

Effects of Melastoma Dodecandrum Preconditioning on Coagulation and Inflammation in Rats with Ischemia Reperfusion Injury of Limb

摘要 目的:研究地稔水煎液预处理对肢体缺血再灌注损伤大鼠的凝血功能及相关炎症指标的影响。方法:30只SD大鼠随机分为对照组、模型组、治疗组,每组10只。治疗组术前5 d予以地稔水煎液连续灌胃处理,1次/d,对照组、模型组两组予等量生理盐水灌胃。采用无创动脉夹夹闭股动脉结合橡皮筋捆扎大腿根部的方法建立大鼠下肢缺血再灌注模型,缺血4 h后恢复灌注,再灌注4 h后取血,全自动血凝分析仪检测各组大鼠凝血酶原时间(PT)、活化部分凝血活酶时间(APTT)、纤维蛋白原(Fib)及血浆凝血酶时间(TT);ELISA法检测细胞间粘附分子1(ICAM-1)、白细胞介素6(IL-6)和肿瘤坏死因子α(TNF-α)的情况。结果:与对照组比较,模型组PT(8.99±0.64)s、TT(18.84±3.11)s和APTT(19.55±3.74)s缩短,Fib含量(2.05±0.24)g/L增加(P<0.05);ICAM-1(728.41±96.68)pg/mL、IL-6(89.04±7.31)pg/mL及TNF-α(38.85±3.15)pg/mL含量则增高(P<0.05)。与模型组比,治疗组Fib(1.76±0.21)g/L、ICAM-1(603.05±44.09)pg/mL、IL-6(76.50±6.92)pg/mL及TNF-α(30.85±4.33)pg/mL显著降低(P<0.01)。治疗组TT(22.45±3.23)s和APTT(23.97±4.56)s延长,与模型组比较差异有统计学意义(P<0.05),其中PT(10.03±0.56)s延长更明显(P<0.01)。结论:肢体缺血再灌注损伤可致大鼠血液处于高凝状态,纤溶功能受抑,并且发生明显的炎症反应。地稔水煎液的预处理可改善上述大鼠凝血功能的紊乱,同时可以减少血液中炎症因子的表达,进而具有一定的抗炎作用。 Objective To investigate the effects of Melastoma Dodecandrum preconditioning on coagulation and inflammation in rats with ischemia reperfusion injury of limb.Methods Thirty Sprague-Dawley rats were randomly divided into three groups: control group(CG, n=10), ischemia-reperfusion group(IR, n=10) and Melastoma Dodecandrum treatment group(MD, n =10).At 5 d before the operation, the Melastoma Dodecandrum decoction was given continuous gavage once daily.The limb ischemia reperfusion injury model was established by clamping femoral artery and binding the thigh root for 4 h in these animals.The Fib, PT, APTT, TT, ICAM-1, IL-6 and TNF-α contents in serum of rats were determined.Results Compared with the CG group, PT(8.99±0.64) s, TT(18.84±3.11) s and APTT(19.55±3.74) s were significantly shortened(P〈0.05) and the concentration of Fib(2.05±0.24) g/L, ICAM-1(728.41±96.68) pg/mL, IL-6(89.04±7.31) pg/mL and TNF-α(38.85±3.15) pg/mL was significantly higher(P〈0.05) in IR group.However, compared with the IR group, Melastoma Dodecandrum significantly decreased the levels of Fib(1.76±0.21) g/L, ICAM-1(603.05±44.09) pg/mL,IL-6(76.50±6.92)pg/mL and TNF-α(30.85±4.33) pg/mL(P〈0.01) and prolonged APTT(23.97 ±4.56) s, TT(22.45±3.23) s(P〈0.05), prolonged PT(10.03±0.56) s(P〈0.01).Conclusion Melastoma Dodecandrum pretreatment has inhibitive effects on blood hypercoagulability and low fibrinolysis state in limb ischemia-reperfusion rats, also reducing the expression of inflammatory cytokine, which shows protective effect on limb ischemia-reperfusion injury in rats.
