UHPLC法同时测定肝康灵颗粒中5个苷类成分

摘要 目的:建立超高压液相色谱(UHPLC)法同时测定肝康灵颗粒(赤芍、栀子、虎杖、田基黄等)中栀子苷、白芍苷、芍药苷、虎杖苷、异槲皮苷的含量。方法:色谱柱:Shim-pack XR-ODSII C_(18)(2.0 mm×75 mm,1.7μm);流动相:乙腈(A)-0.1%磷酸溶液(B)梯度洗脱;检测波长:230 nm;流速:0.3 mL/min,进样体积:1.0μL,柱温25℃。结果:栀子苷、白芍苷、芍药苷、虎杖苷、异槲皮苷分别在0.7~35μg/mL(r=0.9999)、0.5~25μg/mL(r=0.9999)、7~350μg/mL(r=0.9998)、5.5~275μg/mL(r=0.9999)、3~150μg/mL(r=0.9999)范围内线性关系良好,平均回收率分别为100.14%、99.02%、98.81%、98.35%、100.04%,RSD分别为0.54%、1.99%、0.66%、0.84%、0.98%。结论:该方法简便、快速、准确可靠,可为肝康灵颗粒的质量控制提供依据。 Objective: To establish an ultra high performance liquid chromatography( UHPLC) method for the simultaneous determination of gardenoside,albiflorin,paeoniflorin,polydatin and isoquercitrin in Gankangling Granules( Paeonia veitchii Lynch,Gardenia jasminoides Ellis,Polygonum cuspidatum Sieb. et Zucc.,Hypericum japonicum Thunb. etc.). Methods: The analysis of dilute ethanol extract of this drug was performed on Shim-pack XR-ODSII C_(18)( 2. 0 mm × 75 mm,1. 7μm),mobile phase was acetonitrile( A)-0.1 % formic acid( B) in a gradient elution manner,detection wave length was set at 230 nm,injection volume was 1. 0 μL,and column temperature was maintained at 25℃. Results: Paeoniflorin,gardenoside,albiflorin,polydatin and isoquercitrin displayed good linear relationships within the ranges of 0. 7 ~ 35 μg/mL( r = 0. 9999),0. 5 ~ 25 μg/mL( r = 0. 9999),7 ~ 350 μg/mL( r = 0. 9998),5. 5 ~ 275μg/mL( r = 0. 9999) and 3 ~ 150μg/mL( r = 0. 9999),and the average recoveries were 100. 14 %,99. 02 %,98. 81 %,98. 35 % and100. 04 % with the RSDs of 0. 54 %,1. 99 %,0. 66 %,0. 84 % and 0. 98 %,respectively. Conclusion: The method is simple,rapid,accurate,which can be applied as a standard for the quality control of Gankangling Granules.
