HPLC法同时测定两种抗宫炎软胶囊中连翘酯苷B和金石蚕苷的含量

Determination of the Active Ingredient Content in Kanggongyan Soft Capsule Prepared with Two Matrixes by HPLC

目的:建立同时测定用脂溶性和水溶性基质制备的抗宫炎软胶囊中有效成分连翘酯苷B和金石蚕苷的含量测定方法。方法:采用高效液相色谱法,Thermo Hypersil C18色谱柱(250mm×4.6mm,5μm),流动相为乙腈-0.2%磷酸溶液(14∶86);流速0.8 m L/min;检测波长332 nm;柱温30℃。结果:连翘酯苷B与金石蚕苷对照品进样量分别在0.02443~2.03575μg(r=0.9999)、0.02400~2.00002μg(r=0.9999)范围内呈良好的线性关系,平均加样回收率为103.3%(RSD=1.8%)、96.2%(RSD=0.4%)。所测8批样品连翘酯苷B和金石蚕苷含量分别为1.6744~2.9924mg/g、2.2271~3.4162 mg/g。结论:该方法简便、稳定、重复性好,可为抗宫炎软胶囊质量控制提供依据。

Objective： To develop a HPLC method to simultaneously determine active ingredients forsythiaside B and poliumoside in Kanggongyan soft capsule prepared with water-soluble and fat-soluble matrixes. Methods： Thermo Hypersil C18（ 250 mm × 4. 6 mm,5 μm） was used with the mobile phase of acetonitrile-0. 2% phosphoric acid（ 14∶ 86） at the flow rate of 0. 8 m L/min,the detection wavelength was 332 nm,and the column temperature was at 30℃. Results： The linear range of forsythiaside B and poliumoside were 0. 02443-2. 03575μg（ r = 0. 9999） and 0. 02400-2. 00002μg（ r = 0. 9999）. The average recoveries（ n = 6） were 103. 3%（ RSD = 1. 8%） and 96. 2%（ RSD = 0. 4%） respectively. The content of the two ingredients within eight samples determined were 1.6744 ~ 2. 9924 mg/g and 2. 2271 ~ 3. 4162 mg/g. Conclusion： The method was simple,stable with good repeatability and can be used for the quality control of Kanggongyan soft capsule.
