局部高碳酸与高脂肪酸对HCC827移植瘤钙化和生长的影响

Effect of local high fatty acid and high carbon acid on tumor calcification and growth in an HCC827 xenograft model of human lung adenocarcinoma

摘要 目的探讨局部高脂肪酸和高碳酸对HCC827人肺腺癌移植瘤钙化及生长的影响。方法选取40只直径在10~15 mm之间的HCC827人肺腺癌移植瘤裸鼠模型,随机分为4组,对照组,油酸组,碳酸氢钠(NaHCO_3)组,油酸+NaHCO_3组,每组10只,分别瘤内注射0.9%NaCl 0.2 ml,油酸0.2 ml,NaHCO_3(595 mmol/L)0.2 ml,油酸0.1 ml+NaHCO_3(595 mmol/L)0.1 ml,观察肿瘤生长的体积大小及肿瘤组织钙盐沉积情况,结果用SPSS 17.0统计软件对肿瘤体积变化进行分析,肿瘤体积大小用均数±标准差表示,多组间比较采用方差分析,各组间两两比较采用LSD-t检验。结果各组移植瘤注药后体积从小到大依次是油酸组、油酸+NaHCO_3组、NaHCO_3组和对照组,组间差异有统计学意义(F=5.773,P=0.002<0.01);注药后体积差值组间差异有统计学意义(F=7.242,P=0.001<0.01)。由于分组误差,所以各组在注药前体积大小不一,但差异无统计学意义(P>0.05)。各组间两两比较的LSD-t检验结果显示,与对照组比较,油酸组、NaHCO_3组、油酸+NaHCO_3组注药后体积及体积差值差异均有统计学意义(注药后体积:P分别为0.001、0.006、0.002,体积差值:P分别为0、0.047、0.002);与油酸组比较,NaHCO_3组、油酸+NaHCO_3组注药后体积及体积差值差异均有统计学意义(注药后体积:P分别为0.420、0.742,体积差值:P分别为0.023、0.275);与NaHCO_3组比较,油酸+NaHCO_3组注药后体积及体积差值差异均有统计学意义(注药后体积:P为0.631,体积差值:P为0.212)。HE染色结果显示:油酸组和油酸+NaHCO_3组明显有钙盐沉积,NaHCO_3组和对照组未见明显钙盐沉积。结论局部高油酸可促进肿瘤钙盐沉积,油酸可抑制肿瘤的生长;局部高NaHCO_3可抑制肿瘤生长,但停用NaHCO_3后肿瘤体积又会继续增长,可能与NaHCO_3改变了肿瘤的偏酸性环境有关,并且油酸和NaHCO_3有协同抑制肿瘤生长的趋势。 Objective To investigate the effect of local high fatty acid and high carbon acid on tumor calcification and growth in an HCC827 xenograft model of human lung adenocarcinoma. Methods Forty HCC827 human lung adenocarcinoma-bearing nude mice with an average tumor diameter of 10-15 mm were chosen and randomly divided into four groups to receive intratumor injection of oleic acid alone, sodium bicarbonate alone, oleic acid plus sodium bicarbonate, and normal saline, respectively. The changes of tumor volume and calcium deposition were then observed. Results HE staining results showed that obvious calcium salt deposition occurred in the oleic acid alone group and oleic acid + sodium bicarbonate group, but not in the sodium bicarbonate alone group and normal saline group, which suggested that local high fatty acid promotes tumor calcification. At the end of the experiment, the oleic acid alone group had the least tumor volume, followed by the oleic acid + sodium bicarbonate group, sodium bicarbonate alone group, and normal saline group, suggesting that oleic acid can inhibit tumor growth. Local high sodium bicarbonate inhibited tumor growth, but tumor volume continued to grow after stopping using sodium bicarbonate Conclusion Local high oleic acid can promote tumor calcium salt deposition, and oleic acid can inhibit tumor growth. Local high sodium bicarbonate inhibits tumor growth, which may be partially associated with changing the tumor acid environment by sodium bicarbonate.
