胆管癌中长链非编码RNASOX2OT表达水平与调控作用及临床相关性分析 被引量:10

摘要 目的分析lncRNA-SOX2OT在胆管癌组织和细胞中的相对表达水平,探讨RNA对细胞增殖、迁移以及侵袭能力的影响。方法选取对数生长期胆管癌细胞并利用Lipofectamine 2000转染干扰RNA(si-SOX2OT、si-NC)。采用qRT-PCR法检测lncRNA-SOX2OT表达量,MTT实验观察培养24、48、72、96 h细胞增殖能力,Transwell实验观察细胞迁移、侵袭能力。结果胆管癌与癌旁组织相比,其SOX2OT表达明显升高(P<0.05),且SOX2OT上调表达与患者淋巴结侵犯(P=0.031)、TNM分期(P=0.007)、术后复发(P=0.002)有相关性。胆管癌细胞(RBE,QBC939)与正常胆管细胞(HIBEC)相比,其SOX2OT高表达,且si-SOX2OT转染可有效降低胆管癌细胞的增殖、迁移和侵袭能力。结论 lncRNA-SOX2OT在胆管癌中高表达,并可有效促进癌细胞的增殖、迁移以及侵袭,且SOX2OT有望成为胆管癌的潜在生物学标志物。 Purpose To evaluate the expression level of long noncoding RNA (lncRNA) SOX2OT in cholangiocarcinoma tissues, and to explore its regulation effects on cell proliferation, migration and invasion. Methods The cholangiocarcinoma cell lines were selected for logarithmic growth phase and transfected with small interfering RNAs (si-SOX2OT, si-NC) by using Lipofectamine 2000. The expression of lncRNA-SOX2OT was detected by qRT-PCR and cell proliferation capacity at 24, 48, 72, 96 hours was evaluated by MTT assay. The transwell experiment was performed to observe cell migration and invasion ability. Results Compared to the adjacent normal tissues, the expression of SOX2OT in eholangiocarcinoma tissues was significantly elevated ( P 〈 0. 05 ) , and the overexpressed SOX2OT was correlated with lymph node invasion ( P = 0. 031 ) , TNM stage ( P = 0. 007 ) and postoperative recurrence of patients. Compared with normal bile duct cell (HIBEC), the expression in BTC cell lines (RBE, QBC939 ) was relative higher and si- SOX20T transfection effectively reduced the proliferation, migration and invasion of cholangiocarcinoma cells. Conclusion lncRNA-SOX20T is highly expressed in cholangiocarcinoma and effectively promote the proliferation, migration and invasion of tumor cells, and SOX20T is expected to be one potential biomarker for cholangiocarcinoma patients.
