靶向干扰ARTN对体外培养Ishikawa细胞增殖及侵袭能力的影响 被引量:1

摘要 目的观察靶向抑制ARTN对子宫内膜癌Ishikawa细胞增殖及侵袭能力的影响。方法构建靶向干扰ARTN的短发夹RNA(pG PU6/GFP/Neo-ARTN shRNA),利用脂质体法转染Ishikawa细胞。采用qRT-PCR及Western blot法检测转染后ARTN mRNA及蛋白的表达。应用CCK法观察细胞增殖能力,Transwell小室实验观察细胞侵袭能力的变化。结果qRT-PCR和Western blot结果显示,pshRNA-A干扰组与pshRNA-NC空载体对照组、空白对照组相比,其ARTN mRNA及蛋白相对表达量均明显减少(F=47.882,P<0.05;F=726.323,P<0.05)。CCK结果显示pshRNA-A干扰组细胞增殖能力显著降低(P<0.05)。Transwell小室侵袭实验结果显示抑制ARTN表达后,pshRNA-A干扰组穿膜细胞数明显减少(F=55.609,P<0.05)。结论 ARTN的短发夹RNA可有效抑制ARTN在Ishikawa细胞中的表达,并显著减弱细胞增殖能力,降低细胞侵袭能力。干扰ARTN有望为子宫内膜癌的靶向治疗提供新的研究方向。
