转录因子GATA3对Kaposi肉瘤相关疱疹病毒感染细胞增殖和凋亡的影响

Effects of Transcription Factor GATA3 on the Proliferation and Apoptosis in Kaposi's Sarcoma-associated Herpesvirus( KSHV) Infected Cells

摘要 目的观察转录因子GATA3对Kaposi肉瘤相关疱疹病毒(Kaposi's sarcoma-associated herpesvirus,KSHV)感染细胞增殖和凋亡的影响。方法培养KSHV感染细胞i SLK-BAC与KSHV未感染细胞i SLK-Puro,采用实时定量PCR(real-time PCR)法和Western blot检测GATA3的表达情况;分别用CCK8法和流式细胞术检测过表达GATA3后细胞的增殖能力与凋亡水平。结果 GATA3在i SLK-BAC细胞中的mRNA和蛋白表达水平均低于其在i SLK-Puro细胞中的表达水平;过表达GATA3后i SLK-BAC细胞增殖能力下降,凋亡率升高。结论转录因子GATA3能够抑制KSHV感染细胞系i SLK-BAC的增殖,并促进其凋亡。 Objective To observe the effects of transcription factor GATA3 on the proliferation and apoptosis in Kaposi's sarcoma-associated herpesvirus(KSHV)infected cells. Methods KSHV infected cells iSLK-BAC and KSHV uninfected cells iSLK-Puro were cultured,and the expression of GATA3 was detected by real-time PCR and Western blot. Using CCK8 assay and flow cytometry to detect the proliferative ability and apoptosis level after GATA3 plasmid was transfected into iSLK-BAC cells.Results The expression level of GATA3 mRNA and protein in iSLK-BAC cells was lower than that in iSLK-Puro cells. After GATA3 plasmid was transfected into iSLK-BAC cells,the proliferative ability was remarkably inhibited and the apoptosis rate increased.Conclusion Transcription factor GATA3 could inhibit the proliferation of KSHVinfected iSLK-BAC cells and promote apoptosis.
