针刺法对卒中后抑郁模型大鼠额叶皮质单胺递质和脑源性生长因子的影响 被引量:8

Effect of Acupuncture on Monoamine Neurotransmitter and Brain-derived Growth Factor in Frontal Cortex of Model Rats with Post-stroke Depression

摘要 目的:观察蒙医针刺对卒中后抑郁(post-stroke depression,PSD)模型大鼠额叶皮质的单胺递质和脑源性神经营养因子(brain derived neurotrophic factor,BDNF)的影响。方法:采用左侧颈总动脉永久性结扎制作脑缺血模型,叠加实施慢性不可预见性刺激和孤养建立PSD模型。成功造模后,实施28 d干预。针刺组(每日1次15 min斜刺相关穴位,深度0.15~0.20 mm),药物组(每日1次经胃灌注4.06 mg·kg^(^(-1))的盐酸氟西汀),空白组(不做任何干预),模型组(每日1次经胃灌注4.06 mg·kg^(^(-1))的蒸馏水)。检测大鼠额叶皮质5-羟色胺(5-hydroxytryptamine,5-HT)、去甲肾上腺素(norepinephrine,NE)、BDNF的水平。结果:与空白组[(13.05±3.23、30.42±12.62、0.36±0.04)g·L^(-1)]比较,模型组大鼠的前额叶皮质5-HT、NE、BDNF[(8.34±3.69、16.83±12.98、0.29±0.07)g·L^(-1)]水平明显降低(P<0.01或P<0.05)。与模型组大鼠比较,针刺组[(12.68±2.76、30.02±9.45、0.36±0.05)g·L^(-1)]和药物组[(12.81±3.35、29.17±12.95、0.36±0.05)g·L^(-1)]大鼠前额叶皮质5-HT、NE、BDNF的水平明显增加(P<0.05)。针刺组前额叶皮质5-HT、NE、BDNF水平和药物组比较,差异无统计学意义(P>0.05)。结论:蒙医针刺法可能通过调解前额叶皮质5-HT、NE、BDNF的水平起到抗抑郁作用。 Objective:To observe the effect of Mongolian acupuncture on monoamine neurotransmitters and brain-derived neurotrophic factor( BDNF) in frontal cortex of rats with post-stroke depression( PSD). Methods:Model of cerebral ischemia was established by permanent ligation of the left common carotid artery and superimposed with chronic unpredictable stimulation and isolation to establish a PSD model. After successful modeling,28 days of intervention was implemented. Acupuncture group(1 time per day with 15 minutes of oblique stabbing related points,depth 0. 15-0. 20 mm),drug group( daily once perfusion of fluoxetine hydrochloride 4. 06 mg·kg-1),blank group(without any intervention),model group(distilled water with gastric perfusion of4. 06 mg·kg-1 once daily). The levels of 5-hydroxytryptamine(5-HT),norepinephrine( NE),and BDNF in the frontal cortex of rats were measured. Results:Compared with the blank group [(13. 05 ± 3. 23,30. 42 ± 12. 62,0. 36 ± 0. 04) g·L-1],the prefrontal cortex 5-HT,NE,and BDNF in the model group [8. 34 ± 3. 69,16. 83 ± 12. 98) g·L-1,(0. 29 ± 0. 07) g·L-1]levels decreased significantly( P〈0. 01 or P〈0. 05). Compared with model rats,acupuncture group [12. 68 ± 2. 76,30. 02 ± 9. 45 g·L-1,(0. 36 ± 0. 05) g·L-1]and drug group[12. 81 ± 3. 35,29. 17 ± 12. 95]The levels of 5-HT,NE,and BDNF in the prefrontal cortex of rats with g·L-1 and(0. 36 ± 0. 05) g·L-1 increased significantly( P〈0. 05). There was no significant difference in the level of 5-HT,NE,BDNF in the prefrontal cortex between the acupuncture group and the drug group( P〈0. 05). Conclusion:Mongolian acupuncture may play an antidepressant effect by mediating the levels of 5-HT,NE,and BDNF in the prefrontal cortex.
