接骨丸中不同剂量士的宁和马钱子碱对大鼠肝肾功能的影响 被引量:4

Effects of Different Doses of Strychnine and Brucine on Liver Function and Renal Function in Rats

摘要 目的:观察接骨丸中不同剂量士的宁和马钱子碱对大鼠肝肾功能的影响。方法:40只健康大鼠,随机选择10只作为空白对照组;其余30只作为实验组,分为接骨丸低剂量组、接骨丸高剂量组和接骨丸生马钱子组,每组各10只。实验组大鼠给予不同剂量接骨丸灌胃,空白对照组给予等量蒸馏水灌胃。12周后,检测血常规:红细胞(red blood cell count,RBC)、白细胞(white blood cell,WBC)、血红蛋白(hemoglobin,HB)、血小板(platelets,PLT)、中性粒细胞百分比、淋巴细胞百分比、凝血时间(clotting time,CT),血液生化指标:谷丙转氨酶(alanine transaminas,ALT)、谷草转氨酶(aspartate aminotransferase,AST)、碱性磷酯酶(alkaline phosphatase,ALP)、血糖(glucosuria,GLU)、三酰甘油(triacylglycerol,TG)、尿素氮(blood urea nitrogen,BUN)、肌酐(creatinine,Cr)、总胆红素(total bilirubin,T-BIL)、白蛋白(albumin,ALB)。停药2周后取各脏器测定脏器系数。结果:各组大鼠PLT、RBC、WBC和CT比较,差异均无统计学意义(P>0.05);与接骨丸低剂量组比较,接骨丸高剂量组和接骨丸生马钱子组血红蛋白、中性细胞粒百分比显著下降(P<0.05);与接骨丸低剂量组比较,接骨丸高剂量组和接骨丸生马钱子组,淋巴细胞百分比显著上升(P<0.05);接骨丸高剂量组、接骨丸生马钱子组超氧化物歧化酶(superoxide dismutase,SOD)、丙二醛(malonaldehyde,MDA)和肿瘤坏死因子(tumor necrosis factor-α,TNF-α)与空白对照组比较,差异有统计学意义(P<0.05);各组AST、ALP、GLU、T-BIL、BUN、Cr、TG和ALB比较,差异均无统计学意义(P>0.05);接骨丸高剂量组和接骨丸生马钱子组ALT水平明显提高(P<0.05);各组大鼠心脏、脾脏、肺脏、大脑脏器系数比较,差异无统计学意义(P>0.05);与空白对照组比较,接骨丸高剂量组和接骨丸生马钱子组肝脏、肾脏系数明显升高(P<0.05)。结论:标准质量生产的接骨丸中士的宁和马钱子碱剂量的不同对大鼠肝肾功能影响不同,当剂量大于0.4 g·kg^(^(-1))后,对大鼠肝肾功能损伤较大。 Objective:To observe the effects of different doses of strychnine and brucine on liver and kidney function in rats.Methods:Forty healthy rats were randomly selected as blank control group;the remaining 30 were used as experimental group,which was divided into low dose group of Jiegu Pill,high dose group of Jiegu Pill,and Qianzi group of Jiegu Pills,with 10 rats in each group.Rats in the experimental group were given different doses of Jiegu Pills by gavage,and the control group was given equal volume of distilled water by gavage.After 12 weeks,blood tests were performed:red blood cell count(RBC),white blood cell count(WBC),hemoglobin(HB),platelets(PLT),neutrophil percentage Percentage of lymphocytes,clotting time(CT),blood biochemical indicators:alanine transaminas(ALT),aspartate aminotransferase(AST),alkaline phosphatase(ALP),blood glucose(glucosuria,GLU),triacylglycerol(TG),blood urea nitrogen(BUN),creatinine(Cr),total bilirubin(T-BIL),albumin,ALB).After 2 weeks of drug withdrawal,organs were taken to measure the organ coefficient.Results:PLT,RBC,WBC and CT in all groups were not statistically significant(P〈0. 05).Compared with that of the low dose group of Jiegu Pill,the high dose group of Jiegu Pill and the Qigu Pills were used to produce hemoglobin and neutrophils.The percentage of granules decreased significantly(P〈0. 05).Compared with the low dose group of Jiegu Pills,the percentage of lymphocytes in the high dose group of Jiegu Pill and the Shengzi Qianzi group of Jiegu Pills increased significantly(P〈0. 05).The levels of superoxide dismutase(SOD),malonaldehyde(MDA),and tumor necrosis factor-α(TNF-α)in the Maqianzi group and the high dose group of Jiegu Pills were significantly different from that of the control group(P〈0. 05).There was no significant difference in AST,ALP,GLU,T-BIL,BUN,Cr,TG and ALB between groups(P〈0. 05).The ALT level in the high dose group of Jiegu Pill and Maqianzi group of Jiegu Pill was significantly increased(P〈0. 05).There was no significant difference in the coefficient of heart,spleen,lung,and viscera between the rats in each group(P〉0. 05).The kidney coefficient in high-dose group of Juegu Pills and Maqianzi group of Jiegu Pill was significantly increased(P〈0. 05).Conclusion:Different doses of Strychnine and Brucine in the Jiegu Pills with standard quality have different effects on liver and kidney function in rats.When the dose is greater than 0.4 g·kg-1,the liver and kidney function of rats would be greatly damaged.
