摘要
目的探讨参附汤治疗AngⅡ导致心肌细胞肥大的机制。方法建立AngⅡ致心肌细胞肥大模型,采用MTT法、CCK-8试剂盒检测心肌细胞的存活率,测定搏动频率、采用rt PCR测定相关基因Caspase-3、Beclin1和Atg5的表达量。结果与空白组相比,AngⅡ组细胞的存活率降低,搏动频率增加,相关基因Caspase-3、Beclin1和Atg5的表达量明显升高;参附汤含药血清及血中移行成分组可使AngⅡ致损的乳鼠心肌细胞的存活率升高,降低细胞搏动频率和抑制Caspase-3、Beclin1和Atg5的表达。结论参附汤对AngⅡ导致的心肌肥大细胞具有保护作用,其机制是通过抑制细胞自噬和凋亡。
Objective To explore the mechanism of Shenfu decoction in the treatment of cardiac myocyte hypertrophy induced by Ⅱ. Methods Adriamycin was used to establish the model of cardiac myocyte hypertrophy induced by AngⅡ, the survival rate of cardiomyocytes was detected by MTT assay and CCK-8 kit, and the expression of Caspase-3, Beclinl and Atg5 of related genes was determined by rtPCR. Results Compared with the blank group, the cell survival rate of AngⅡ group decreased, the pulse frequency increased, and the expression of related genes Caspase-3, Beclinl and Atg5 increased significantly. The drug-containing serum of Shenfu decoction and the Transitional component group in the blood can increase the survival rate of neonatal rat cardiomyocytes damaged by AngⅡ, reduce the frequency of cell beats and inhibit the expression of Caspase-3, Beclinl and Atg5. Conclusion Shenfu decoction has protective effect on myocardial mast cells induced by angii, and its mechanism is to inhibit autophagy and apoptosis of cells.
