罗格列酮对家兔颅脑损伤周围脑组织的保护作用

Protective effects of rosiglitazone on perifocal brain tissue after traumatic brain injury in rabbits

摘要 目的观察微创穿刺手术后穿刺轨迹周围脑组织过氧化物酶体增殖物激活受体γ(peroxisome proliferator activated receptorγ,PPARγ)水平、过氧化氢酶(catalase,CAT)表达及脑组织含水量的变化,探讨灌注罗格列酮对颅脑损伤脑组织的保护作用。方法将45只健康雄性家兔随机分为对照组、损伤组和罗格列酮组,每组15只。家兔在颅脑损伤模型制作成功后,对照组不行微创穿刺手术,损伤组及罗格列酮组模拟微创穿刺手术,随后罗格列酮组穿刺轨迹区灌注罗格列酮。术后7 d,RTPCR法检测3组损伤周围脑组织PPARγ mRNA及CAT mRNA的表达,测量损伤周围脑组织含水量。结果损伤组PPARγ mRNA及CAT mRNA表达高于对照组(P<0.05);与损伤组比较,罗格列酮组PPARγ mRNA及CAT mRNA表达显著增加(P<0.05)。与对照组比较,损伤组损伤区周围脑组织含水量显著增加(P<0.05);与损伤组比较,罗格列酮组含水量显著降低(P<0.05)。结论微创穿刺手术后穿刺轨迹区灌注罗格列酮能有效减轻家兔颅脑损伤,这种保护作用可能与增加PPARγ mRNA及CAT mRNA的表达、降低脑水肿有关。 Objective To observe expressions of peroxisome proliferator activated receptor γ(PPARγ) and catalase(CAT) and the change of water content of brain tissues around the puncture trajectory after minimally invasive surgery for investigating the protective effects of rosiglitazone on brain tissue after traumatic brain injury(TBI) in rabbits. Methods A total of 45 rabbits were randomly divided into control group, injury group and rosiglitazone group and each group had 15 rabbits. TBI model was prepared on all rabbits successfully. The rabbits of control group did not receive the minimally invasive surgery and rabbits of the other groups received the surgery. And rosiglitazone was infused to the brain tissues around the puncture trajectory in rosiglitazone group. The expressions of PPARγ mRNA and CAT mRNA were detected by RT-PCR and the water content of injury tissues was measured 7 d after surgery.Results Expressions of PPARγ mRNA and CAT mRNA were higher in the injury group than in control group(P〈0.05). Compared with injury group, the expressions of PPARγ mRNA and CAT mRNA more significantly increased in rosiglitazone group(P〈0.05).Compared with control group, the water content of brain tissues around the injury areas was higher in injury group(P〈0.05). The water content was lower in rosiglitazone group than in the the injury group(P〈0.05). Conclusions The rosiglitazone infusion therapy in puncture trajectory areas could relieve the injury of brain tissues of rabbits after minimally invasive surgery, and the protective effects may have relation with the up-regulation of PPARγ mRNA and CAT mRNA and the down-regulation of brain edema.
