骨性关节炎中lncRNA MEG3的表达及其与VEGF的相关性分析 被引量:9

摘要 目的检测长链非编码RNA母系表达基因3(lncRNA MEG3)和血管内皮生长因子(VEGF)在骨性关节炎(OA)软骨组织中的表达情况,并分析lncRNA MEG3表达与VEGF表达之间的相关性。方法 2014年1月~2015年12月,收集新疆维吾尔自治区中医医院40例原发性OA患者和20例志愿者的正常膝关节软骨组织;番红O对软骨组织进行染色后,改良Mankin评分标准对OA患者进行分组;实时定量荧光PCR试验检测各膝关节软骨组织中lncRNA MEG3和VEGF m RNA表达水平,酶联免疫吸附测定(ELISA)试验检测VEGF蛋白水平。Pearson相关系数法分析lncRNA MEG3表达与VEGF表达之间的相关性。结果 Real-time PCR结果显示,与正常软骨组织相比,lncRNA MEG3在OA患者软骨组织中相对表达量显著降低(P<0.05),且软骨退化程度越高,lncRNA MEG3相对表达量越低(P<0.05)。VEGF m RNA和VEGF蛋白相对表达量在OA患者软骨组织中均显著高于正常软骨组织(P<0.05),且软骨退化程度越高,VEGF m RNA和VEGF蛋白相对表达量越高(P<0.05)。Pearson相关分析结果显示,lncRNA MEG3表达与VEGF表达之间呈负相关(r=-0.466,P<0.05)。结论 lncRNA MEG3在OA软骨组织中低表达,且与VEGF表达之间呈负相关,提示lncRNA MEG可能通过调节血管生成参与原发性OA的发生过程。 Objective To determine the expressions of long non-coding RNA MEG3(lncRNA MEG3) and vascular endothelial growth factor(VEGF) in the articular cartilage tissues of osteoarthritis(OA),and to analyze the correlation between the lncRNA MEG3 and VEGF.Methods The knee articular cartilage tissues from 40 cases of primary OA and20 healthy volunteers were collected from January 2014 to December 2015 in Traditional Chinese Medicine Hospital of Xinjiang Uygur Autonomous Region.After the safranin O staining,the OA patients were grouped according to the improved score criterion of Mankin.Real-time PCR assay was applied to detect the expressions of lncRNA MEG3 and VEGF m RNA in the knee articular cartilage tissues,and the ELISA assay was utilized to determine the protein level of VEGF.The correlation between the lncRNA MEG3 and VEGF was analyzed by the Pearson.Results The real-time PCR results showed that compared to the normal articular cartilage tissues,the lncRNA MEG3 was significantly downregulated in the OA articular cartilage tissues(P〈0.05),and the higher degree of degradation,the lower of lncRNA MEG3(P 0.05).Moreover,the m RNA and protein levels of VEGF were significantly upregulated in the OA articular cartilage tissues(P 0.05),and the higher degree of degradation,the higher of VEGF(P 0.05).The Pearson results showed that there was a negative relationship between the lncRNA MEG3 and VEGF(r =-0.466,P 0.05).Conclusion lncRNA MEG3 was decreased in the articular cartilage tissues of OA,and lncRNA MEG3 was negatively correlated with the VEGF.lncRNA MEG3 may be involved in primary OA development through the regulation of angiogenesis.
