微波消解-电感耦合等离子体质谱法测定指甲中锰 被引量:2

Determination of manganese in fingernail by microwave digestion-inductively coupled plasma mass spectrometry

摘要 目的建立用微波消解-电感耦合等离子体质谱法(ICP-MS)快速、准确测定人体指甲中微量元素锰的检测方法。方法称取适量样品经HNO3体系微波消解处理后,以质量数72锗作为内标物质,采用电感耦合等离子体质谱法直接测定样品消解液中锰元素的含量,对职业暴露人群和对照人群的指甲进行了检测分析。结果本法锰元素线性范围在0μg/kg^500μg/kg,标准曲线线性关系良好,相关系数r=0.999 9,方法检出限为0.013μg/g,标准物质GBW07601a(GSH-1a人发)的测定结果在标准参考值范围内,相对标准偏差(RSD)为4.0%,样品3个不同浓度的加标回收率为84.0%~104.6%,样品检测结果暴露组指甲锰的含量远远高于对照组。结论本法灵敏准确、快速方便,适用于人体指甲中微量锰元素的测定分析,可以为评估人群中重金属微量锰元素研究工作提供技术支持。 Objective To establish a method for the determination of manganese in fingernail by microwave digestion-inductively coupled plasma mass spectrometry( ICP-MS). Methods The sample was digested with HNO3 microwave and manganese content was determined by ICP-MS with germanium 72 as internal standard ; the fingernails of the occupational exposure population and the control group were detected and analyzed. Results Good linearity was obtained when the concentration of manganese was within 0 μg/kg-500 μg/kg, with correlation coefficient(r) of 0.999 9, and the detection limit of the method was 0.013 μg/g. The determination result of the standard substance GBW07601a (GSH-la hair) was within the standard reference range, the relative standard deviation(RSD) was less than 4.0% , and the spiked recovery, of three concentrations was within 84. 0%-104. 6%; the manganese content in exposed group was much higher than that in the control group. Conclusion The method is sensitive, accurate, simple and rapid, and it can be applied to the determination of manganese in fingernail.
