日落黄荧光光谱测定方法与量子产率的研究

摘要 应用荧光光谱法研究食用合成色素日落黄的荧光光谱特性.在p H=9的磷酸盐缓冲溶液的实验条件下,λex/λem=260/376 nm,研究了金属离子、食用酸、氧化还原剂、糖类、防腐剂等对日落黄荧光光谱的影响.在此基础上建立了测定日落黄的荧光光谱分析法,线性回归方程:IF=11.24C+56.06,线性范围:1.0~7.0 mg/L,相关系数r=0.9959,检出限0.174 mg/L.测定了醒目饮料(菠萝味)中日落黄的含量,7次测量结果的相对标准偏差RSD=2.37%,同时,测定出样品中日落黄的回收率在93%~102%之间,能定量回收.以硫酸奎宁为参比,测量了日落黄在不同波长下的荧光量子产率在0.1~0.7之间. Fluorescence spectrometry is applied in this article to study the fluorescence spectral characteristics of edible synthetic pigment yellow sunset(sunset yellow,FCF). Metal ions,food acids,pigment,oxidant,reducing agent,carbohydrate,preservatives,heavy atom,quencher and ethanol medium's different influences on sunset yellow fluorescence have been studied under the experimental condition of Phosphate buffer solution on λex/λem =260/376 nm,p H =9. On this basis,it established Fluorescence Spectroscopy to determine Sunset Yellow. Among which,linear regression equation is IF=11.24 C +56.06,the linear range being 1.0 ~7.0 mg/L,the correlation coefficient r =0.9959,and the detection limit is 0.174 mg/L. This method is also used to determine the content of sunset yellow in striking drinks which tastes as pineapple. After seven measurements,the relative standard deviation RSD equals 2.374%. At the same time,the recovery of sunset yellow in the sample is determined,ranging from 93% to102%,which leads to the result that sunset yellow can be recovery quantitatively. Using quinine bisulphate as a reference,fluorescence quantum yields of sunset yellow at different wavelengths are measured,ranging from 0.1 to 0.7.
