蒲参胶囊对大鼠心肌缺血再灌注损伤的保护作用

The protective effect of pushen capsule in rats with myocardial ischemia/reperfusion injury

目的:研究蒲参胶囊对大鼠心肌缺血再灌注损伤的保护作用以及其作用机制。方法:将雄性SD大鼠随机分为假手术组、模型组、复方丹参滴丸阳性药组(600 mg/kg),蒲参胶囊(10、20、40mg/kg)高、中、低剂量组,给予相应药物12周后对大鼠进行心肌缺血再灌注实验,即缺血30min,再灌注45min,取血和心脏,HE染色观察大鼠心肌组织病理形态改变,及对血清乳酸脱氢酶(LDH)、肌酸激酶(CK)活性的影响,心肌细胞超氧化物歧化酶(SOD)活性,丙二醛(MDA)含量、大鼠白细胞介素6(IL-6)和肿瘤坏死因子-α(TNF-α)水平的影响。结果:与假手术组相比,模型组血清LDH、CK活性升高(P<0.05),心肌细胞MDA含量、IL-6和TNF-α水平升高(P<0.05),SOD活性下降(P<0.05);与模型组相比,蒲参胶囊低中高剂量组血清LDH、CK活性显著下降(P<0.05),心肌细胞MDA含量、IL-6和TNF-α水平显著下降(P<0.05),SOD活性显著升高(P<0.05),组织病理学也证实蒲参胶囊对大鼠心肌缺血再灌注损伤有一定的保护作用,降低心肌纤维变性坏死,降低炎性反应。结论:蒲参胶囊对大鼠心肌缺血再灌注损伤具有一定的保护作用,其保护作用可能与提高心肌氧化应激能力,改善心肌细胞炎性反应,维持心肌细胞完整性有关。

Objective：To study the effects of Pushen capsule in mice with myocardial ischemia/reperfusion injury.Methods：Male mice were randomly divided into sham group,model group,salvia dropping pills group and Pushen capsule low-dose,medium-dose and high-dose groups（10,20 and 40 mg/kg）with 6 mice in each group.Those groups were given relevant medicine before modeling for 12 w,once a day.IM/IR model was induced in those groups except for sham operation group.The activities of serum LDH、CK,activity of SOD、MDA、IL-6 and TNF-αin myocardial cells were detected and histopathological changes were determined.Results：Compared with sham group,serum LDH、CK,content of MDA、IL-6 and TNF-αincreased significantly（P〈0.05）in model group while SOD decreased significantly（P〈0.05）.Compared with model group,serum LDH、CK,content of MDA、IL-6 and TNF-αdecreased significantly（P〈0.05）in Pushen capsule groups while SOD increased significantly（P〈0.05）.Histopathology of mice in model group showed Pushen capsule can better the myocardial ischemia/reperfusion injury.Conclusion：Pushen capsule can better the myocardial ischemia/reperfusion injury.The mechanism of that may be associated with decrease oxidative stress and low inflammatory response.
