NEDD9表达变化与神经胶质瘤的恶性表型及不良预后的关系

Relationship between NEDD9 expression and malignant phenotype and prognosis of gliomas

目的:探讨NEDD9表达变化与神经胶质瘤的恶性表型及不良预后的关系。方法:选择神经胶质瘤患者100例,根据恶性程度分为低度恶性组(Ⅰ~Ⅱ级胶质瘤)和高度恶性组(Ⅲ~Ⅳ级胶质瘤),每组各50例。同时收集重度颅脑外伤患者50例作为对照组。采用免疫组化法检测NEDD9在各组脑组织中的表达情况;采用qRT-PCR和Western blot法检测各组脑组织中NEDD9 mRNA和蛋白表达情况,随访5年观察术后两组胶质瘤患者预后情况,分析NEDD9蛋白表达与预后的关系。结果:免疫组织化学染色实验结果显示,在对照组的脑组织中NED9蛋白的表达极低,在低度恶性组和高度恶性组中NEDD9表达较高,且高度恶性组明显高于低度恶性组(P<0.05);胶质瘤患者脑组织中NEDD9 mRNA及蛋白相对表达量均高于对照组(P<0.05);术后NEDD9蛋白低表达组生存率明显高于NEDD9蛋白高表达组(P<0.05);多因素COX回归分析显示,NEDD9蛋白高表达是神经胶质瘤患者生存期缩短的独立危险因素。结论:NEDD9表达与神经胶质瘤的恶性表型关系密切,与神经胶质瘤恶性进展有关,是患者一个独立的预后危险因素。

Objective：To investigate the relationship between the expression of NEDD9 and the malignant phenotype and prognosis of gliomas. Methods：100 patients with glioma were selected,according to the degree of malignancy,these patients were divided into two groups,low malignant group（ grade Ⅰ - Ⅱ grade） and high malignant group（ grade Ⅲ - Ⅳ grade）,each group was 50 cases. At the same time,50 patients with severe traumatic brain injury were collected resected as control group. Immunohistochemistry was used to detect the expression of NEDD9 in various brain tissues. The expression of NEDD9 mRNA and protein in each group were detected by RT-PCR and Western blot. The prognosis of the two groups were observed after 5-year follow-up. The relationship between NEDD9 protein expression and prognosis was analyzed. Results：The results of immunohistochemical staining showed that NED9 expression was very low in the normal control group,and it was higher in the low malignant group and the high malignant group,NED9 expression in the high malignant group were higher than high malignant group（ P〈0. 05）. The relative expression of NEDD9 mRNA and protein in glioma group were higher than those in normal control group（ P〈0. 05）. Survival rate of NEDD9 low expression group was significantly higher than that of NEDD9 high expression group after operation（ P〈0. 05）. Cox regression analysis showed that high expression of NEDD9 protein was an independent risk factor for the shortened survival of patients with glioma. Conclusion：The expression of NEDD9 is closely related to the malignant phenotype of glioma,and is related to malignant progression of glioma. It is an independent prognostic risk factor for glioma.
