摘要
目的观察健胃愈脑汤对脑缺血再灌注大鼠认知功能的影响。方法选取30只成年雄性SD大鼠为研究对象,随机分为对照组、假手术组和健胃愈脑汤组各10只,将假手术组和健胃愈脑汤组大鼠制成脑缺血再灌注模型,健胃愈脑汤组大鼠术后1d灌服健胃愈脑汤,其余2组同时灌服等量生理盐水,术后第21~25天行Morris水迷宫检查,评估大鼠认知功能的变化;术后第26天行Real time-PCR和Western blot检测BDNF mRNA和蛋白表达。结果随着大鼠训练时间的延长,潜伏期时间缩短,与假手术组相比,对照组潜伏期时间明显增长(P<0.05),大鼠空间学习能力受损严重;与对照组相比,健胃愈脑汤组潜伏期时间明显缩短(P<0.05),大鼠空间学习能力明显恢复。此外,健胃愈脑汤组大鼠在目标象限停留时间和圆台穿越次数明显多于对照组(P<0.05),说明大鼠空间记忆能力得到显著改善。BDNF mRNA和蛋白检测结果显示,对照组对比于假手术组,BDNF mRNA和蛋白水平均明显下降(P<0.05),而健胃愈脑汤组比对照组BDNF mRNA和蛋白表达水平有明显升高(P<0.05)。结论健胃愈脑汤可明显改善脑缺血再灌注大鼠的认知功能,提高海马区脑源性神经营养因子的表达,促进脑损伤的恢复。
Objective To observe the effect of Jianwei Yunao decoction on the cognitive function of cerebral ischemia-reperfusion rats.Methods Thirty adult male Sprague-Dawley rats were randomly divided into control group,sham operation group and Jianwei Yunao decoction group,10 rats in each group.Rats in sham operation group and Jianwei Yunao decoction group were made Cerebral ischemia-reperfusion model,Jianwei Yunao decoction group rats were given Jianwei Yunao decoction 1 day after operation,the other two groups were given the same amount of saline at the same time,Morris water maze examination was performed on the 21-25 days after operation The changes of cognitive function in rats were evaluated.Real-time PCR and Western blot were used to detect the expression of BDNF mRNA and protein on the 26 th day after operation.Results Compared with the sham-operated group,the latency of the control group increased significantly(P〈0.05)and the spatial learning ability of the rats was severely impaired with the prolongation of training time in rats.Compared with the control group,The incubation period of Jianwei Yunao decoction group was significantly shortened(P〈0.05),and the spatial learning ability of rats was obviously recovered.In addition,the rats in Jianwei Yunao decoction group had significantly more dwell time in the target quadrant and the number of roundabout traversing than that in the control group(P〈0.05),indicating that the spatial memory ability of rats was significantly improved.The results of BDNF mRNA and protein showed that the BDNF mRNA and protein levels in the control group were significantly decreased compared with the sham operation group(P〈0.05),while the expression of BDNF mRNA and protein in the Jianwei Yunao decoction group was significantly higher than that in the control group(P〈0.05).Conclusion Jianwei Yunao decoction can obviously improve the cognitive function of cerebral ischemia-reperfusion rats,increase the expression of brain-derived neurotrophic factor in the hippocampus and promote the recovery of brain injury.
