黄芪甲苷对氯吡格雷所致胃黏膜损伤大鼠血清IL-6、IL-8、TNF-α的影响

Effects of Astragaloside Ⅳ on Serum IL-6,IL-8 and TNF-α in Rats with Gastric Mucosal Injury Induced by Clopidogrel

目的探讨黄芪甲苷对氯吡格雷所致大鼠胃黏膜损伤的保护作用及血清IL-6、IL-8、TNF-α的影响。方法健康雄性SD大鼠50只,随机分为对照组、模型组及黄芪甲苷低、中、高剂量组,每组10只,分别予生理盐水、氯吡格雷、氯吡格雷联合不同浓度黄芪甲苷灌胃,每日1次,7d后处死大鼠。Guth法测定胃黏膜溃疡指数;ELISA法检测血清IL-6、IL-8、TNF-α水平;光镜下观察大鼠黏膜病理形态变化。结果与对照组相比,模型组大鼠胃黏膜损伤指数升高,差异有统计学意义(P<0.01);与模型组相比,各实验组大鼠胃黏膜损伤指数改善,差异有统计学意义(P<0.05);模型组血清IL-6、IL-8、TNF-α水平明显高于对照组,差异有统计学意义(P<0.05);与模型组相比,各实验组血清IL-6、IL-8、TNF-α水平降低,差异有统计学意义(P<0.05)。结论黄芪甲苷可以通过降低血清炎性因子IL-6、IL-8、TNF-α水平对氯吡格雷所致胃黏膜损伤发挥保护作用。

Objective To investigate the protective effect of astragaloside Ⅳ on clopidogrel-induced gastric mucosal injury in rats and the effects on serum IL-6,IL-8 and TNF-α.Methods Fifty healthy male Sprague-Dawley rats were randomly divided into control group,model group and low,medium and high dose astragaloside groups（10 rats for each group）.Saline,clopidogrel and clopidogrel combined with different concentration of astragaloside IV was intragastrically administered in each group once a day and the rats were sacrificed 7 days later.The gastric mucosal ulcer index was measured by Guth method;the levels of serum IL-6,IL-8,and TNF-αwere determined by ELISA;the pathological changes of the mucosa were observed under light microscope.Results Compared with the control group,the indices of gastric mucosal injury in the model group increased with significant difference（P〈0.01）.Compared with the model group,the gastric mucosal injury indices were improved in the experimental group,and the difference was statistically significant（P〈0.05）;serum levels of IL-6,IL-8 and TNF-αin the model group were significantly higher than those in the control group,and the difference was statistically significant（P〈0.05）;compared with the model group,serum levels of IL-6,IL-8 and TNF-αin each experimental group decreased and the difference was statistically significant（P〈0.05）.Conclusion Astragaloside IV can protect clopidogrel-induced gastric mucosal injury by decreasing the levels of serum inflammatory factors IL-6,IL-8 and TNF-α.
