摘要
目的研究桂籽中环烯醚萜苷的提取纯化以及含量测定方法。方法采用95%乙醇回流提取,石油醚(60℃~90℃)、乙酸乙酯、水饱和正丁醇依次萃取;以特女贞苷、女贞苷G13为对照品,采用紫外全波长扫描(UV)、高效液相色谱法(HPLC)对桂籽环烯醚萜苷进行含量测定。结果特女贞苷、女贞苷G13在24.72μg/m L^494.41μg/m L、24.50μg/m L^490.00μg/m L线性关系良好,回归方程分别为Y=4.6045X-21.392,R2=0.9996;Y=3.9901X-10.508,R2=0.9992。桂籽正丁醇萃取部位中特女贞苷和女贞苷G13的百分含量分别为39.20%、39.88%,经大孔树脂纯化后,特女贞苷、女贞苷G13进一步富集,含量分别为82.56%、87.94%。结论桂籽环烯醚萜苷的提取方法简单方便,提取率较高,含量测定方法稳定、重现性好,可用于桂籽环烯醚萜苷的含量测定。
Objective To explore the extraction,purification and content determination of the iridoid glycosides in Osmanthus fragrans seeds. Methods Firstly,95% ethanol was used for reflux extraction. Secondly,the iridoid glycosides was enriched by extraction refined through petroleum ether( 60℃ ~ 90℃),ethyl acetate and water saturated of n-butanol sequentially. Specnuezhenide and Nuezhenoside G13 was used as the control products. The iridoid glycosides in Osmanthus fragrans were determined by UV and HPLC. Results Specnuezhenide and Nuezhenoside G13 are in good linear relationship within 24. 72μg/m L-494. 41μg/m L,24. 50μg/m L-490. 00μg/m Lseparately. Regression equations are Y = 4. 6045 X-21. 392,R2= 0. 9996; Y =3. 9901 X-10. 508,R2= 0. 9992. The content of the Specnuezhenide and Nuezhenoside G13 in the n-butanol extracts of Osmanthus fragrans seeds were 39. 20% and 39. 88% respectively. Actually,their content reached up to 82. 56% and 87. 94% after purified by macroporous resin. Conclusion The extraction method is simple and convenient,and the extraction rate is high. The method is stable and reproducible. It can be used to examine the content of the iridoid glycosides of Osmanthus fragrans seeds.
