抗植物病原线虫的多粘类芽胞杆菌KM2501-1发酵培养基和发酵条件优化 被引量:5

Optimization of Fermentation Culture Medium and Fermentation Conditions of Paenibacillus polymyxa KM2501-1 Against Phytopathogenic Nematodes

摘要 多粘类芽胞杆菌KM2501-1可以有效抑制南方根结线虫对植物的危害,促进植物生长,具有良好的应用前景。采用正交实验和单因素实验分别对多粘类芽胞杆菌KM2501-1的发酵培养基和发酵条件进行了优化。结果表明,多粘类芽胞杆菌KM2501-1发酵培养基的最优配方为:可溶性淀粉3%、豆粕3%、氯化钙0.05%、氯化钠0.25%;最适发酵初始pH值为8.5,最适发酵温度为28℃。将多粘类芽胞杆菌KM2501-1在优化培养基中的发酵上清液稀释5倍后,作用根结线虫48 h的校正死亡率为87.15%,比优化前提高了61.61%;多粘类芽胞杆菌KM2501-1在优化培养基中发酵48 h后的芽胞数为2.5×10~8 cfu·mL^(-1),是优化前的6.25倍,芽胞率达到100%时发酵周期为48 h,优化效果十分显著。该研究为多粘类芽胞杆菌KM2501-1的中试和大规模发酵生产提供了基础数据。 Paenibacillus polymyxa KM2501-1 can effectively inhibit the damage to plants caused by Meloi- dogyne incognita , promote plant growth,and has good application prospects. We optimized the fermentation cul-ture medium and fermentation conditions by an orthogonal ex periment and a single-factor experiment, respec-tively. The results show that the optimum formulation of fermentation culture medium for Paenibacillus poly- myxa KM2501-1 is as follows: 3% soluble starch,3% soybean meal,0. 05% calcium chloride,and 0. 25% sodi-um chloride. The optimum initial fermentation pH value is 8. 5 , and the optimum fermentation temperature is 28 C. After 5-fold dilution of the fermentation supernatant of Paenibacillus polymyxa KM2501-1 in the opti-mized culture medium , t he nematode correction mortality rate after 48 h exposing is 87. 15% , which is 61. 61%higher than the rate before optimization. The spore number of KM 2501-1 is 2. 5 X 108 cfu ?times the number before optimization. W h e n the sprouting rate reaches 1 0 0% , the fermentation ph , showing that the optimization efficiency is very significant. The study provides tscale and large-scale fermentation production of Paenibacillus polymyxa KM 2501-1.
