摘要 目的建立同时测定甘草及其炮制品中甘草苷和甘草酸含量的超快速液相色谱法,为甘草药材及其炮制品的质量控制提供参考。方法采用Shim-Pack XR-ODS柱(75 mm×3.0 mm,2.2μm);流动相为0.05%磷酸水(A)-乙腈(B),梯度洗脱(0~4 min,5%~20%B;4~12 min,20%~40%B;12~15 min,40%~45%B),平衡时间为2 min;流速为0.8 m L/min;检测波长为237 nm;柱温为30℃。结果甘草苷和甘草酸分别在10~500 mg/L(R=0.999 8)、10~500 mg/L(R=0.999 9)浓度范围内与峰面积呈良好的线性关系;平均回收率分别为98.9%和99.3%。结论该方法能够准确地测定甘草中甘草苷与甘草酸的含量,操作简便,具有良好的重现性和稳定性,可用于甘草药材的质量控制。 Objective To establish an ultra-fast liquid chromatography( UFLC) method for simultaneous determination of glycyrrhizin and glycyrrhizic acid in Glycyrrhiza uralensis Fisch. and its processed products,and provide references for its quality control. Methods Chromatographic separation was performed on Shim-Pack XR-ODS column(75 mm × 3. 0 mm,2. 2 μm). The mobile phase consisted of 0. 1% formic acid(A)-acetonitrile(B) with gradient elution(0 - 4 min,5% - 20% B;4 - 12 min 20% - 40% B;12 - 15 min,40% - 45% B) at a flow rate of 0. 8 mg/L and the equilibration time was 2 min. The detecting wavelength was set at 237 nm and column temperature was maintained at 30 ℃. Results The linear ranges were 10 - 500 mg/L( R = 0. 999 7) for glycyrrhizin and 10 - 500 mg/L( R =0. 999 8) for glycyrrhizic acid. The average recovery of glycyrrhizin and glycyrrhizic acid were 98. 9% and 99. 3%,respectively. Conclusion The developed method is simple,accurate with high repeatability and stability,which is suitable for the quality control of Glycyrrhiza uralensis Fisch.
