直链烷基苯磺酸钠对雌性小鼠卵巢类固醇激素合成的影响 被引量:3

Effect of Linear Alkylbenzene Sulfonate on Synthesis of Steroid Hormone in Ovaries of Female Mice

摘要 目的探讨直链烷基苯磺酸钠(LAS)对雌性小鼠卵巢类固醇激素合成的影响。方法将40只妊娠小鼠随机分为研究组和对照组,各20只。研究组于妊娠第5~20天以150 mg/(kg·d)LAS灌胃染毒,对照组小鼠同期每天以等量生理盐水灌胃。两组均正常分娩,观察其子代雌性小鼠发育情况至10周龄,在子代雌性小鼠发情间期处死,用酶联免疫吸附(ELISA)法检测并比较两组子代雌性小鼠血清卵泡刺激素(FSH)、黄体生成素(LH)、雌二醇(E2)及睾酮(T)等类固醇激素水平,血清超氧化物歧化酶(SOD)活力及丙二醛(MDA)等氧化应激指标水平,并对其卵巢中类固醇激素急性调节蛋白(St AR)、细胞色素P450胆固醇侧链裂解酶(P450scc)mRNA表达量进行检测和比较。结果子代雌性小鼠研究组13只,对照组15只;与对照组子代雌性小鼠比较,研究组子代雌性小鼠的血清类固醇激素水平、血清SOD活力及卵巢St AR和P450scc mRNA相对表达量均降低,血清MDA水平则升高(P<0.05);Pearson线性相关分析结果显示,研究组子代雌性小鼠血清SOD活力与其血清类固醇激素水平及卵巢St AR和P450scc mRNA相对表达量均呈正相关(r=0.866,0.805,0.798,0.836,0.818,0.897,P<0.05),其血清MDA水平与其血清类固醇激素水平及卵巢St AR和P450scc mRNA相对表达量则呈负相关(r=-0.811,-0.835,-0.872,-0.818,-0.806,-0.857,P<0.05)。结论 LAS可影响雌性小鼠卵巢类固醇激素合成,而氧化应激可能为LAS影响卵巢类固醇激素合成的机制。 Objective To investigate the effect of Linear Alkylbenzene Sulfonate(LAS) on synthesis of steroid hormone in ovaries of female mice. Methods Totally 40 pregnant mice were randomly divided into the study group and the control group,20 cases in each group. The study group was given 150 mg/(kg·d) LAS by gavage every day during 5 ~ 20 d of pregnancy,and the control group was given the same amount of normal saline by gavage every day in the same period. The two groups were normal childbirth,and the development of offspring female mice were observed to 10 weeks of age,and then the offspring female mice were killed in diestrus. Enzyme-linked immunosorbent assay(ELISA) method was adopted in the detection and comparison of offspring female mice serum follicle stimulating hormone(FSH),luteinizing hormone(LH),estradiol(E2) and testosterone(T) and other steroid hormone levels,and serum superoxide dismutase(SOD) activity,malondialdehyde(MDA) and other oxidative stress indicators levels in the two groups. The ovarian steroid acute regulatory protein(St AR) and cytochrome P450 cholesterol side chain cleavage(P450 scc) mRNA expression were detected and compared in the two groups. Results There were 13 offspring female mice in the study group and 15 offspring female mice in the control group. Compared with those of the offspring female mice of the control group,the serum steroid hormone level,serum SOD activity level,ovary St AR and P450 scc mRNA relative expression level of the offspring female mice of the study group were decreased,while the serum MDA level of the offspring female mice of the study group was increased(P〈0. 05). Pearson linear correlation analysis showed that the serum SOD activity level of the offspring female mice of the study group was positively correlated with the serum steroid hormone level and the ovaries St AR and P450 scc mRNA relative expression level(r = 0. 866,0. 805,0. 798,0. 836,0. 818,0. 897,P〈0. 05),while the serum MDA level of the offspring female mice of the study group was negatively correlated with the serum steroid hormone level and the ovaries St AR and P450 scc mRNA relative expression level(r =-0. 811,-0. 835,-0. 872,-0. 818,-0. 806,-0. 857,P〈0. 05). Conclusion LAS can affect the synthesis of steroid hormone in the ovaries of female mice,and oxidative stress may be the mechanism of LAS affecting the synthesis of ovarian steroid hormone.
