摘要
目的探讨miR-135b对亮氨酸拉链肿瘤抑制基因1(LZST1)表达及肺癌细胞转移行为的影响及机制。方法 q PCR检测miR-135b和LZST1在肺癌组织和癌旁正常组织中的表达情况。通过转染miR-135b-mimic和miR-135b-inhibitor模拟物上调和下调肺癌A549细胞中miR-135b的表达。Western blotting检测miR-135b对LZST1的调控作用;划痕愈合实验和Transwell侵袭实验检测miR-135b对肺癌细胞的迁移和侵袭能力的影响;免疫组织化学检测肺癌组织和癌旁正常组织中LZTS1蛋白的表达情况。结果肺癌组织中miR-135b和LZST1表达水平高于癌旁正常组织(P<0.05)。LZST1蛋白在肺癌组织中表达阳性,在癌旁正常组织中表达阴性。与阴性对照组相比,miR-135b-mimic组LZST1蛋白表达上调,miR-135b-inhibitor组LZST1蛋白表达下调(P<0.05)。与阴性对照组相比,miR-135b-mimic组的迁移能力和侵袭能力增强,miR-135b-inhibitor组的迁移能力和侵袭能力减弱(P<0.05)。结论 miR-135b可以通过调控LZST1的表达影响肺癌A549细胞的迁移和侵袭能力。
Objective To investigate effects and its mechanism of miR-135 b on leucine zipper tumor suppressor gene 1( LZST1) expression and metastasis of lung cancer cells. Methods Expressions of miR-135 b and LZST1 in lung cancer tissues and adjacent normal tissues were detected by quantitative polymerase chain reaction( q PCR). Expression of miR-135 b in lung cancer A549 cells was up-regulated and down-regulated by transfecting miR-135 b-mimic and miR-135 b-inhibitor mimics. Western blotting was used to detect regulatory effect of miR-135 b on LZST1. Scratch healing assay and Transwell invasion assay were used to detect effects of miR-135 b on migration and invasion of lung cancer cells.Immunohistochemistry method was used to detect expressions of LZTS1 protein in lung cancer tissues and adjacent normal tissues. Results Expression levels of miR-135 b and LZST1 in lung cancer tissues were higher than those in adjacent normal tissues( P〈0. 05). Expressions of LZST1 protein were positive in lung cancer tissues and negative in adjacent normal tissues. Compared with those in negative control group,expression of LZST1 protein in miR-135 b-mimic group was up-regulated,while the expression in miR-135 b-inhibitor group was down-regulated( P〈0. 05). Compared with those in negative control group,migration and invasion abilities in miR-135 b-mimic group were enhanced,while the abilities in group miR-135 b-inhibitor were weakened( P〈0. 05). Conclusion The miR-135 b may affect migration and invasion abilities of lung cancer A549 cells by regulating expression of LZST1 in lung cancer cell.
