防石汤对大鼠肾草酸钙结石防治效果的实验研究 被引量:5

摘要 目的:研究防石汤对大鼠肾草酸钙结石的防治效果,初步探究其作用机制,为临床用药提供实验依据。方法:将60只SD大鼠随机分为5组,即A组(正常组)、B组(成石组)、C1组(低剂量防石汤组)、C2组(中剂量防石汤组)、C3组(高剂量防石汤组)。用乙二醇法复制大鼠肾草酸钙结石模型,喂养4周后处死,取出双肾后甲醛溶液固定,HE染色石蜡切片,显微镜下观察肾组织内草酸钙晶体分布情况并测定大鼠24 h尿pH值及草酸、钙的含量。用SPSS 24.0统计软件对实验数据进行处理。结果:与A组比较,B组成石率、尿草酸、尿钙升高差异均有显著性(P<0.05);与B组相比,C1、C2、C3组成石率、尿草酸下降差异均有显著性(P<0.05);与C2组比较,C1、C3组成石率、尿草酸、尿钙差异均有显著性(P<0.05);C1与C3组成石率、尿草酸、尿钙相比,差异均无显著性(P>0.05);各组尿pH值相比差异均无显著性(P>0.05)。结论:防石汤对大鼠肾草酸钙结石治疗效果确切,以中剂量组疗效最佳。 Objective: To study the preventive and therapeutic effect of Fang Shi Tang on calcium oxalate stone in rats,and to explore its mechanism of action,so as to provide experimental evidence for clinical medication. Methods: 60 SD rats were randomly divided into 5 groups,namely,group A(normal group),group B(stone group),group C1(low dose of Fang Shi Tang group),C2 group(medium dose group),and C3 group(high dose group). The model rats of calcium oxalate stone were replicated by ethylene glycol. After feeding for 4 weeks,the rats were sacrificed. The double kidneys were taken out and fixed with formaldehyde solution,and the paraffin section was stained with HE. The distribution of calcium oxalate crystals in the kidney tissue was observed under microscope and the pH value of 24 h urine and the content of oxalic acid and calcium were measured. The experimental datas were processed with SPSS 24. 0 statistical software. Results: Compared with group A,the difference in the stone rate,urinary oxalic acid and urine calcium of group B was significant(P 〈 0. 05). Compared with group B,the stone rate and urine oxalic acid of C1,C2,C3 decreased significantly(P 〈 0. 05). Compared with group C2,the difference in the stone rate,urinary oxalic acid and urinary calcium of group C1 and C3 was significant(P 〈 0. 05). There was no significant difference in stone formation rate,urinary oxalate and urine calcium between group C1 and group C3(P 〉 0. 05),and there was no significant difference in urine pH value between each group(P〉 0. 05). Conclusion: Fang Shi Tang has definite effect on renal calcium oxalate stone in rats,and the best effect is in the medium dosage group.
