摘要
目的通过Meta分析,评价细胞色素P450 1A1(CYP1A1)基因Ile462Val多态性与宫颈癌易感性的关系。方法计算机检索PubMed、Medline、CNKI及万方数据库,收集国内外已发表有关CYP1A1基因Ile462Val多态性与宫颈癌相关性的病例—对照研究,检索时限均为建库至2017年9月,对入选文献以OR为效应指标,采用Stata1 2.0软件对符合纳入标准的研究进行Meta分析。结果最终纳入14篇文献,共2 223例宫颈癌病例和2 424例健康对照;Meta分析结果显示,CYP1A1基因Ile462Val多态性与宫颈癌发病风险相关性具有统计学意义[OR=1.673,95%CI=1.205~2.321,P=0.002];按种族分层后,高加索人群:[OR=2.335,95%CI=1.544~3.531,P=0.000];亚洲人群:[OR=1.274,95%CI=0.844~1.992,P=0.249。]结论 CYP1A1基因Ile462Val的多态性增加高加索人群宫颈癌发病风险,但与亚洲人群宫颈癌的发病风险无明显相关性。受纳入研究数量及质量限制,该结论仍需进一步验证。
Objective To evaluate the relationship between Cytochrome P450 1A1( CYP1A1) Ile462 Val polymorphism and cervical cancer by meta-analysis. Methods The case-control studies about the correlation between CYP1A1 Ile462 Val polymorphism and cervical cancer were retrieved from Pub Med,Medline,CNKI and Wan Fang Data base. The retrieval time was from their establishment to September,2017. After the paper screened,data extracted,and bias risk assessed,the meta analysis was conducted by Stata 12. 0 software. Results A total of 14 case-control studies involving 2 223 cervical cancer patients and 2 424 controls were included. The results of meta-analysis showed a significant association between the CYP1A1 Ile462 Val polymorphism and cervical cancer risk[OR = 1. 673,95% CI = 1. 205 - 2. 321,P =0. 002]. Stratification analysis by ethnicity showed significant association in the population of Caucasian origin[OR = 2. 335,95% CI = 1. 544 - 3. 531,P = 0. 000 ],but not in Asian origin[OR = 1. 274,95% CI =0. 844 - 1. 922,P = 0. 249]. Conclusion To date,the current evidence shows that the CYP1A1 Ile462 Val polymorphism may be associated with cervical cancer risk in Caucasian,but not in Asians. For the quantity and quality limitation of the included studies,the conclusion has to be verified by more large-scale high quality studies.
