摘要
目的:观察番石榴叶提取物对自发2型糖尿病db/db小鼠肾脏HE染色及TGF-β1免疫组化的影响,探讨其肾脏保护的机制。方法:10周龄雄性肥胖db/db小鼠16只,适应性喂养2周后,随机分为模型组和番石榴叶提取物组,所有动物给予高脂饲料喂养,番石榴叶提物组按照0.11g/kg灌胃(相当于生药材2g/kg),模型组给予同体积蒸馏水灌胃,疗程为12周。每4周观察体重、血糖变化,试验结束后,HE染色观察肾脏病理变化,免疫组化法观察TGF-β1蛋白表达。结果:与对照组相比,经过12周的治疗后,番石榴叶提取物显著降低db/db小鼠体重和血糖。HE染色结果显示,番石榴叶提取物可以显著改善db/db小鼠肾小球系膜细胞增生、球旁器增生,颗粒样变性。免疫组化结果提示,番石榴叶提取物可以显著下调db/db小鼠肾脏TGF-β1蛋白表达(P<0.05)。结论:番石榴叶提取物通过抑制肾脏组织TGF-β1免疫组化蛋白表达,改善db/db小鼠糖尿病肾病肾脏纤维化。
Objective:To observe treatment of Guava leaf extracts on renal tissue TGF-β1 protein expression in db/db mice.Methods:16 male obese db/db mice,10 weeks old,were randomly divided into control group and Guava leaf water extracts(0.11 g/kg)after being fed for 2 weeks.All mice received high fat diet and 12-week treatment.Body weight and glucose were tested every week.HE staining according to the manufacturers' instructions.Renal tissue TGF-β1 protein was tested by immunohistochemistry,The relative levels of each target protein to controlβ-actin were determined by densitometry analysis using the Image-Pro Plus.Results:GL extracts significantly lowered the values of body weight and plasma glucose,reduced the proliferation of glomerular mesangial cells,proliferation of peripheral organs,and granular degeneration.Furthermore,treatment with GL extracts decreased renal tissue TGF-β1 protein expression by immunohistochemistry.Conclusion:GL extracts may improve fibrosis by decrease TGF-β1 signaling pathway in renal tissue.
