摘要 以绿豆皮为原料,主要研究了绿豆皮黄酮的纯化工艺。首先比较了AB-8,LSA-10,HPD-100和ADS-7型4种大孔树脂对绿豆皮黄酮的吸附和解析特性,选择其中最好的树脂,对其吸附和解析条件进行了研究。结果表明,AB-8型是纯化绿豆皮黄酮较好的大孔树脂,其最佳吸附条件为上样速率1.98 m L/min,上样质量浓度0.045 9 mg/m L,上样量10 m L时,最大吸附率可达88.89%;其最佳解析条件为解析液乙醇体积分数70%,解析液洗脱流速1.98 m L/min,解析液用量25 m L时,解析率可达69.51%。绿豆皮黄酮的纯度由10.57%提高至72.19%,表明该纯化工艺具有较好的纯化效果,可为绿豆皮黄酮的纯化及进一步开发应用提供参考。 In this paper, the purification technology of flavonoids from Mung tegument was studied. Firstly, the adsorption and desorption properties of macroporous resins such as AB-8, LSA-10, HPD-100 and ADS-7 were compared. The preferred resin was screened and the adsorption and analytical conditions were studied. The results showed that AB-8 was the best macroporous resin with the best flavonoids. The best adsorption conditions were as follows: the loading rate was 1.98 mL/min, adsorption liquid concentration of 0.045 9 mg/mL, the upper sample was 10 mL, the maximum adsorption rate could up to 88.89%. Its best analytical conditions: analytical liquid concentration of 70%, the flow rate was 1.98 mL/min, and analytical liquid dosage was 25 mL, the analytical rate was 69.51%. The purity of flavonoids in the extract was increased from 10.57% to 72.19%, which indicated that the purification process had better purification effect, which can provide reference for the purification and further development and application of flavonoids from Mung tegument.
