响应面法优化缬草多糖的提取工艺及其抗氧化性

Optimizing of Extraction Process of Polysaccharides from Valeriana officinalis L via Response Surface Method and Its' Antioxidant Activity

摘要 采用响应面法优化缬草多糖的提取工艺,并测定其体外抗氧活性。在单因素试验基础上,考察提取温度、提取时间和料液比对缬草多糖得率的影响。结果表明:在提取时间2.6h、提取温度81℃、料液比1:27(g:m L)条件下,多糖得率最高,为17.00%,与理论值17.24%相差较小;不同浓度多糖(0.2~1 mg/m L)对DPPH(62.9%~82.5%)和羟基自由基(45.6%~80.2%)都有较好的清除效果,且在一定范围内与浓度呈正相关性。由此可知,所得回归模型可用于缬草多糖提取工艺预测,且缬草多糖在体外具有较好的抗氧化活性。 The extracting process of the polysaccharides from Valerian root was optimized by using response surface experients. At the same time, the anti-oxidant activity of polysaccharide was measured by anti-oxidant tests. Based on single factor tests, the impact of temperature, time, and ratio of materials on the yield of polysaccharides extracted from Valerian root was analyzed. The results showed that the optimazed processing is as follows, extracting time 2.6 h, solid-liquid 1:27(g:m L) and temperature 81℃. Under the optimal condition, the heighest yield of the polysaccharides was 17.00%, which was close to the theoretical value of 17.24%(Relative deviation was 0.24%). With concentrations of the Valerian polysaccharides ranging from 0.2 to 1 mg/ml, it had signifcant effect on the scavenging rate of DPPH(62.9%~82.5%)and Hydroxyl radical(45.6%~80.2%), and there was a positive correlation between the scavenging rate and the concentration of the Valerian polysaccharides in a certain range. In conclusion, it was feasible to extract valerian polysaccharides by response surface methodology, and valerian polysaccharide has good antioxidant activity in vitro.
