黄芪-当归配伍对大鼠血管内膜增生模型细胞外基质的影响 被引量:8

Effect of Combination of Astragali Radix and Angelicae Sinensis Radix on Extracellular Matrix of Intimal Hyperplasia Rat Models

摘要 目的研究黄芪和当归配伍对大鼠血管内膜增生血管壁细胞外基质的影响。方法将SD大鼠随机分为黄芪与当归(简称芪归)1∶1、芪归5∶1(3.9 g/kg)配伍组及单用黄芪组(2.17 g/kg)、单用当归组(1.08 g/kg)、阳性药物阿托伐他汀对照组(0.1 g/kg)和假手术组,采用球囊导管损伤大鼠血管内皮造成胸腹主动脉血管内膜增生模型,同时灌胃黄芪、当归不同比例配伍药物,连续给药14 d后,取胸腹主动脉,用western blot法检测增生内膜中基质金属蛋白酶-9(MMP-9)、组织基质金属蛋白酶抑制物-1(TIMP-1)蛋白表达,免疫组化法检测增生内膜中Ⅰ型胶原(Col-Ⅰ)、纤维联接蛋白(FN)表达。结果与模型组比较,单用当归组、芪归1∶1组、阿托伐他汀组Col-Ⅰ、FN、MMP-9表达显著降低(P<0.01,P<0.05)。芪归5∶1组Col-Ⅰ、MMP-9表达强度显著低于模型组(P<0.05);单用黄芪组MMP-9表达强度显著低于模型组(P<0.05)。单用黄芪组Col-Ⅰ、FN的表达强度与模型组比较差异无统计学意义(P>0.05),芪归5∶1组FN的表达强度与模型组比较差异无统计学意义(P>0.05)。各组间TIMP的表达强度差异无统计学意义(P>0.05)。结论黄芪-当归配伍可抑制血管内膜增生时血管壁细胞外基质沉积,其中当归为其主要的效应药物,黄芪-当归1∶1配伍的作用强于两药单用及其5∶1配伍。 Objective To study the effect of compatibility of Astragali Radix (Huangqi) and Angelicae Sinensis Radix(Danggui) on the extracellular matrix of rats with intima hyperplasia. Methods Sprague -Dawley rats were randomly dividedinto different groups: Huangqi-Danggui1:1 group, Huangqi-Danggui5:1 group (3.9 g/kg), Huangqi group (2.17 g/kg), Danggui group(1.08 g/kg), atorvastatin control group (0.1g/kg) and sham-operation group. A model of intimal hyperplasia of thoracoabdominal aortawas established by balloon catheter injury in rats. Then thoracoabdominal aorta was taken out after administration of Huangqi andDanggui for 14 days. The expression of matrix metallopeptidase-9 (MMP-9) and tissue inhibitor of metalloproteinases-1 (TIMP-1) inproliferating endometrium was detected by Western blot. Immunohistochemistry was used to detect the expression of collagen-Ⅰ (Col-Ⅰ) and fibronectin (FN) in proliferative endometrium. Results Compared with the model group, the expression intensity of Col-Ⅰ, FN and MMP-9 in Danggui group, Danggui-Huangqi1:1 group, atorvastatin group were significantly lower (P〈0.01, P〈0.05). Theexpression of Col-Ⅰ and MMP-9 in Danggui-Huangqi5:1 group was significantly lower than that in model group (P 〈0. 05) . The expression ofMMP-9 in Danggui group was significantly lower than that in model group (P〈0.05) . There was no significant difference in the expressionintensity of Col-Ⅰ and FN between the Danggui group and the model group (P〉0.05). There was no significant difference in theexpression intensity of FN between the Huangqi-Danggui5:1 group and the model group(P〉0.05). There was no significant differencein the expression intensity of TIMP between each group(P〉0.05). Conclusion Huangqi and Danggui could inhibit the deposition ofextracellular matrix during intimal hyperplasia. And Danggui is one of the main effect drugs. The effect of Huangqi-Danggui1:1compatibility was stronger than the two drugs alone and compatibility of Huangqi-Danggui5:1.
