摘要
目的研究补肾化瘀方对多囊卵巢综合征(polycystic ovary syndrome,PCOS)模型大鼠血清25羟基维生素D_3[25-(OH)VD_3]、抗苗勒管激素(AMH)、血清睾酮(T)、胰岛素(INS)及雌二醇(E_2)的影响。方法 61只SD雌性大鼠随机选取8只设为空白组,剩余大鼠采用来曲唑灌胃法建立PCOS大鼠模型。成模后大鼠随机分为模型组、中药组及西药组,给予相应药物干预后腹主动脉取血。采用酶联免疫吸附法测定药物干预后各组大鼠血清激素[25-(OH)VD_3、AMH、T、INS、E_2]含量,分析各组血清激素水平的差异。结果空白组、中药组以及西药组的血清E_2与25(OH)VD_3浓度高于模型组,差异有统计学意义(P<0.05或P<0.01),而AMH与INS浓度均低于模型组,差异有统计学意义(P<0.05或P<0.01)。结论补肾化瘀方对灌服来曲唑所致PCOS模型大鼠血清AMH、INS、25(OH)VD_3与E_2等激素水平有调节作用,能有效改善PCOS的无排卵、卵泡发育停滞及糖脂代谢紊乱等问题。
Objective To study the effect of Bushen Huayu decoction on serum vitamin D3, adrenal medullary hyperplasia (AMH), testosterone (T), insulin (INS) and estradiol (E2) in polycystic ovary syndrome (PCOS) rats. Methods Eight rats were randomly selected from 61 SD female as the blank group. The remaining rats by gastric perfusion of letrozole were to establishment of PCOS rat models. After modeling, the rats were randomly divided into the model group, the Chinese medicine group and the Western medicine group. The blood of abdominal aorta was taken after the intervention of the corresponding drug intervention. The serum levels of 25 (OH)VD3, AMH, T, INS, and E2 in each group after drug intervention were measured by enzyme-linked immunosorbent assay (ELISA). The difference of serum levels in each group was analyzed. Results The concentrations of serum E2 and 25 (O H) V D3 in the blank group, the Chinese medicine group and the western medicine group were higher than those in the model group, the results were significantly different (P〈0.05 or P〈0.01). The serum AMH and INS concentrations in the three groups were lower than those in the model group, which were significantly different (P〈0.05 or P〈0.01). Conclusion Bushen Huayu decoction can regulate the levels of AMH, INS, 25(OH)VD3 and E2 in the serum of letrozole-induced polycystic ovary syndrome (PCOS) rat models, which can effectively improve PCOS's anovulatory, follicular development stagnation and disorder of glycolipid metabolism.
