摘要
DNA疫苗进入机体在肌肉细胞、吞噬细胞中表达,直接或间接呈递给T细胞。研究发现,DNA疫苗自身作为一种病原相关分子模式被细胞质内DNA感受器识别、结合,通过激发Ⅰ型干扰素、前炎性细胞因子等启动固有免疫应答从而激活适应性免疫应答,达到了解DNA疫苗的免疫机制改善DNA疫苗的免疫原性的目的。论文通过对国内外关于某些DNA感受器及DNA代谢产物作为分子佐剂机制的最新研究进行归纳,以及将其作为佐剂的试验研究做一总结,为研制活性强、高效低毒、有治疗作用的新型佐剂作为改善DNA疫苗免疫性的靶点提供了参考。
The DNA vaccines get into the body,expres in muscle cells,phagocytic cells,and directly or indirectly present to T cells.Recent studies have found that DNA vaccine itself as a pathogen associated molecular pattern is recognized and bound in the cytoplasm of DNA receptor,and activates innate immune responses and adaptive immune response through activation of type I interferon,proinflammatory cytokines,for understanding the immune mechanism of DNA vaccine to improve the immunogenicity of DNA vaccine.This paper summarized the latest research on some DNA receptors and DNA metabolites as the molecular adjuvant mechanism,as well as the experimental study of adjuvants.Therefore,it provided new ideas for developing novel adjuvants with activty,high efficiency,low toxicity,and therapeutic effect,as a target for improving immunination of DNA vaccine.
