摘要
采用体外法探讨不同浓度丁酸对瘤胃上皮细胞生长及凋亡的影响。不同浓度丁酸(0,20,40,60,80,100,120,140 mmol/L)处理瘤胃上皮细胞不同时间(4,6,8,10 h)后,MTT法检测各处理组的细胞活力,再选择适宜处理用于TUNEL法、DNA ladder法、流式细胞仪检测细胞凋亡情况。结果表明:随着丁酸添加浓度的升高,瘤胃上皮细胞的相对细胞活力先增加后降低,20 mmol/L丁酸能促进瘤胃上皮细胞的生长,40~140 mmol/L丁酸则降低了瘤胃上皮细胞的细胞活力。添加120 mmol/L丁酸培养6 h诱导了瘤胃上皮细胞发生凋亡,总凋亡指数为28.4%,显著(P<0.05)高于对照组(24.1%)。低浓度丁酸可促进瘤胃上皮细胞生长,而高浓度丁酸则促进细胞发生凋亡。
The aim of this study was to investigate the effect of butyric acid on the growth and apoptosis of rumen epithelial cells in vitro. Rumen epithelial cells were treated with different concentrations of butyrate acid(0,20,40,60,80,100,120,140 mmol/L) and different treatment time(4,6,8,10 h). After that,the viability of epithelial cells in each group was detected by MTT assay. Then the suitable treatment group was used to detect cell apoptosis by TUNEL assay,DNA ladder assay and flow cytometry. The results showed that:with the increase of butyric acid concentration,the relative cell viability of rumen epithelial cells increased firstly,and then decreased. Additive of 20 mmol/L butyric acid promoted the growth of rumen epithelial cells,while 40-140 mmol/L butyrate decreased the viability of ruminal epithelial cells.Apoptosis detection revealed that apoptosis of rumen epithelial cells induced by the additive of 120 mmol/L butyric acid cultured for 6 h.The total apoptosis index of epithelial cells in 120 mmol/L butyrate group was 28.4%,which was significantly higher than that in the control group(24.1%,P〈0.05).In conclusion,low concentration of butyric acid promoted the growth rate of rumen epithelial cells,while high concentration of butyric acid induced the apoptosis of rumen epithelial cells.
