针刺百会穴与皮质γ-氨基丁酸能神经元缺血后功能障碍的相关性研究 被引量:7

摘要 目的:探讨电刺激百会穴是否对皮质γ-氨基丁酸(GABA)能神经元的缺血后功能障碍具有保护作用。方法:本研究分为缺血组、缺血加针灸组及对照组,3组小鼠于第20天处死后取脑制备脑片,缺血组及缺血加针灸组经缺血处理后,通过全细胞记录观察3组小鼠皮质GABA能神经元对缺血的反应。结果:与对照组及缺血组相比,缺血加针灸组可以改善大脑皮层GABA能神经元产生群集动作电位容量的降低(P<0.01),降低缺血导致的大脑皮层GABA能神经元群集发放动作电位阈电位的提高和不应期(ARP)的延长(P<0.01),百会组GABA能神经元缺血后突触反应的幅度和频率均显著改善(P<0.01)。结论:针刺百会穴能改善缺血后动作电位损伤,拮抗缺血后阈电位的升高和不应期的延长,保护缺血后皮质GABA能神经元突触反应的损伤,从而改善缺血性卒中的预后。 Objective: To investigate whether electrical stimulation of Baihui( GV20) has a protective effect on cortical GABAergic neurons after ischemia stroke. Methods: The mice were randomly divided into the ischemia group,the ischemia plus acupuncture group and the control group. After ischemia intervention,the responses of cortical GABAergic neurons to ischemia were recorded in the three groups. The three groups of mice were sacrificed on the 20 th day and the brains were taken to prepare brain slices. Results: The ischemia plus acupuncture group could significantly improve the decrease of the action potential of cortical GABAergic neurons,compared to the control group and the ischemic group( P〈0. 01); it regulated the increase of threshold potential and prolongation of refractory period of ischemia-induced aggregation of GABAergic neurons in the cerebral cortex( P〈0. 01); the amplitude and frequency of post-ischemic synaptic responses of GABAergic neurons in the ischemia plus acupuncture group were significantly improved( P〈0. 01). Conclusion: Needling GV20 can improve the injury of action potential,antagonize the increase of the threshold potential and the prolongation of refractory period after ischemia,and protect the cortical GABAergic neurons from the synaptic reaction injury and improve the prognosis of ischemic stroke.
