PM_(2.5)对大鼠脾细胞中氧化损伤及凋亡蛋白的影响 被引量:3

摘要 为探讨大气细颗粒物PM_(2.5)暴露对Wistar大鼠急性脾损伤的影响,PM_(2.5)冬夏混合膜由实验室保留,将大鼠随机分为实验组和对照组(n=8),实验组4 h/d、750μg·mL^(-3)PM_(2.5)口鼻暴露处理,对照组生理盐水,5 d/周,共2周,染尘结束24 h后处死,称重收集脾脏上清和细胞,HE染色观察脾组织形态学变化。检测脾上清H_2O_2含量水平。Cell viabitity assay和qRT—PCR仪器检测脾细胞的Bax、Bc L-2水平的表达情况。HE染色结果发现,PM_(2.5)可增加脾生发中心的淋巴细胞。PM_(2.5)染尘后实验组大鼠体重明显低于对照组,差异均有统计学意义(P<0.05)。PM_(2.5)染尘可增加脾上清H2O2含量水平。脾细胞活力和凋亡结果表明,PM_(2.5)暴露可抑制脾细胞的增殖能力以及上调Bax和Bcl-2基因表达。PM_(2.5)染尘可引起脾脏氧化损伤,促使氧化应激水平上升,发生炎症反应,增加脾细胞的增殖能力,最终对脾产生一定的损害作用。 To investigate the effect of atmospheric PM2.5 exposure on acute spleen injury in Wistar rats PM2.5 winter and summer mixed membrane retained by the laboratory. Rats were randomly divided into experimental group and control group( n = 8). The rats in the experimental group were treated with 5 h/week for 2 weeks at a dose of 750 μg·m L-3 PM2.5,4 h/d. The rats were sacrificed 24 h after the end of dust,and the supernatant of spleen and cells were collected. HE staining was used to observe the morphological changes of spleen. The levels of H2 O2 in spleen were detected. Cell viabitity assay and qRT-PCR were used to detect the expression of Bax and Bc L-2 in spleen cells. HE staining results showed that PM2.5 can increase spleen germinal center of lymphocytes.The body weight of experimental rats in PM2.5-exposed group was significantly lower than that in control group( P 〈0. 05). PM2.5 can increase the level of H2O2 in spleen supernatant. The results of spleen cell viability and apoptosis showed that PM2.5 exposure could inhibit the proliferation of splenocytes and up-regulate the expression of Bax and Bcl-2 genes. PM2.5 can cause oxidative damage of spleen,promote the level of oxidative stress,the inflammatory reaction,increase the ability of spleen cells,and finally produce some damage to the spleen.
