对叶百部药材中四种主要生物碱的分布规律 被引量:4

摘要 目的:了解百部药材中主要生物碱的含量和分布规律,为百部药材的质量控制提供依据。方法:采用反向HPLC-ELSD方法对百部药材中4种主要生物碱金刚大碱、百部新碱、新对叶百部碱和对叶百部碱的含量就行了测定,以0.1%三乙胺-甲醇(43∶57)为流动相,Agilent C18色谱柱(4.6 mm×250 mm,5μm),柱温30℃,流速1m L/min;ELSD漂移管温度为80℃,氮气流速为2.1 m L/min,增益为8。结果:83批百部药材含金刚大碱、百部新碱、新对叶百部碱和对叶百部碱的药材分别有27、48、22和48批,其平均含量分别为1.98、4.75、3.73和5.34 mg/g;含有1~4种生物碱的药材分别有38、28、17和0批次,其中对叶百部碱与其余三种生物碱中任意一种同时出现的概率较高;购于安徽亳州和广州清平药市的药材不含新对叶百部碱。结论:本文建立的含量测定方法简便、准确、可靠;含量测定结果说明4种生物碱在药材中的分布具有趋单性,仅含一种生物碱的药材占45.8%,含2种及以上生物碱的药材占54.2%。本文研究结果对于对照品短缺情况下,对购买百部药材化学成分的预测具有一定的指导意义。 Objective: To understand the distribution of the alkaloids in Baibu(the root of Stemona tuberosa),the contents of the main alkaloids were determined. Methods: A reverse HPLC-ELSD method was established for the determination of croomine,stemoninine,neotuberostemonine and tuberostemonine in Baibu. They were separated on an Agilent Extend C18 column(4. 6 mm × 250 mm,5μm) with a mobile phase of 0. 1% triethylamine-methanol(43∶ 57),a column temperature of 30℃ and a flow rate of1 ml/min. ELSD was set to 80℃ for drift tube,2. 1 ml/min for N2 and 8 for gain. Results: Among the 83 batches of Baibu,the batches containing croomine,stemoninine,neotuberostemonine and tuberostemonine were 27,48,22 and 48,respectively,with an corresponding average content of 1. 98,4. 75,3. 73 and 5. 34 mg/g. There were 38 batches containing one alkaloid,28 batches containing 2 ones,17 batches containing 3 ones and no batches containing 4 ones. Tuberostemonine are readily associated with the other alkaloids. Baibu purchased from Bozhou(Anhui) and Qingping(Guangzhou) didn't contain neotuberostemonine. Conclusion: The established HPLC-ELSD method was simple,accurate,and reliable. The distribution of the alkaloids tended to be single because 45. 8% batches of Baibu contained only1 alkaloid. This study has some guiding significance for the prediction of the chemical constituents of Baibu under the shortage of reference substance.
