抽真空法和甘油冷冻法保存对高效脱胶菌株CXJZU-120活化性能的比较研究

摘要 为了探索一种使高效脱胶菌株能够长期保存且易活化的方法,试验采用抽真空法和甘油冷冻法两种方法对保存了3年或10年的菌株CXJZU-120进行苎麻脱胶,定期测定活化菌液和发酵液中相关活化性能指标。结果表明:甘油冷冻法保存3年的菌株经活化10 h,菌体颜色深浅不一;发酵10.5 h,其活菌数为1.66×10~7cfu/mL,COD值为3.86 g/L,苎麻脱胶失重率为19.53%,只适合短期保存。然而,抽真空法保存3年、10年的两支菌株经活化10 h,活化菌体颜色均较深;发酵10.5 h,其活菌数均可达1.02×10~9cfu/mL,COD值分别为6.02、5.65 g/L,苎麻脱胶失重率均超过27%。因此,抽真空法适合高效脱胶菌株CXJZU-120中、长期保存。 In order to explore a long-term preservation and easily activating method for high efficiency degumming strain,the related activation performance indexes of activated bacteria and fermentation liquid were tested during ramie degumming using the strain CXJZU-120 preserved for 3 years or10 years by the method of vacuum and glycerol cryopreservation. After the 10 h activation of the strain preserved for 3 years by the method of glycerol cryopreservation,the results showed that the strain color was different,the number of viable bacteria was 1. 66 × 10~7 cfu/mL,COD value was 3. 86 g/L,and the loss rate of ramie degels was 19. 53%,which was indicated that the method was only suitable for short-term preservation. However,after the activation of the strain preserved for 3 years and 10 years by the vacuum method,the strain color was deep,the number of viable bacteria was 1. 02 × 10~9 cfu/mL,COD value were 6. 02 g/L and 5. 65 g/L respectively,and the loss rate of ramie degel was over 27%. Therefore,the vacuum method was suitable for the long-term preservation of the strain CXJZU-120.
