摘要
目的通过生物信息学分析研究结直肠癌与正常结肠粘膜间基因表达的差异,找出关键的差异表达基因,从而预测参与结直肠癌发生发展中的关键基因。方法从GEO表达谱数据库中下载表达谱基因芯片数据GSE50760(包括18例结直肠癌样本及配对的正常结肠粘膜组织)。利用Morpheus在线软件对基因芯片GSE50760的数据进行分析,获得在结直肠癌与正常结肠粘膜组织中差异表达的基因,并构建聚类分析热图。利用human protein atlas在线数据库获得差异表达基因在结直肠癌与正常结肠粘膜组织中的免疫组化表达水平。并进一步利用human protein atlas数据库的数据对差异表达基因在结直肠癌患者中的表达高低进行生存分析。结果通过分析GSE50760获得前100个差异表达的基因。利用human protein atlas在线数据库对这100个差异表达基因的免疫组织表达水平进行分析发现,FAM135B基因在结直肠癌组织中的表达比正常结肠粘膜中的表达明显下调。生存分析也提示当FAM135B基因高表达时,结直肠癌患者的预后相对较好。结论 FAM135B可能是结直肠癌中潜在的抑癌基因,其表达下调在结直肠癌的发生发展过程发挥重要作用,并可能成为判断结直肠癌患者预后的重要的生物学指标。
Objective To investigate the differences in gene expression between colorectal cancer and normal colon mucosa by bioinformatics analysis,and to identify key differentially expressed genes,thereby predicting key genes involved in the development of colorectal cancer. Methods The expression microarray data GSE50760(including 18 colorectal cancer samples and matched normal colon mucosal tissues)was downloaded from the GEO expression profile database. The data of the gene chip GSE50760 was analysed by Morpheus to obtain differentially expressed genes between colorectal cancer tissues and normal colon mucosa tissues,and a cluster analysis heat map was constructed. The online database human protein atlas was used to obtain the immunohistochemical expression of the differentially expressed genes in colorectal cancer and normal colon mucosa. The prognostic value of the differentially expressed genes in colorectal cancer was analysed by survival analysis using the data in human protein atlas. Results The top 100 differentially expressed genes were obtained by analyzing GSE50760. The immunohistochemical expression levels of these 100 differentially expressed genes were analyzed by using the data in human protein atlas. It was found that the expression level of FAM135 B was significantly lower in colorectal cancer tissue than that in normal colon mucosa. The survival analysis also suggested that the higher expression level of FAM135 B was associated with better prognosis of patients with colorectal cancer. Conclusion FAM135 B may be a potential tumor suppressor gene in colorectal cancer. The down-regulation of FAM135 B plays an important role in the occurrence and development of colorectal cancer,and may be an important biological indicator for judging the prognosis of colorectal cancer patients.
