摘要
为了探讨柯里拉京对宫颈癌Hela细胞增殖与凋亡的影响,不同浓度柯里拉京作用Hela细胞后,利用MTT法和克隆形成实验检测柯里拉京对Hela细胞的增殖抑制作用,用倒置显微镜观察Hela细胞的形态变化,用流式细胞仪分析细胞周期与细胞死亡率,用Western blot法检测细胞PCNA、Caspase-3、Caspase-8、Caspase-9、PARP蛋白的表达。结果:柯里拉京能显著抑制宫颈癌Hela细胞增殖,并呈浓度依赖效应;流式细胞仪测定结果表明柯里拉京能使Hela细胞发生S期周期阻滞;Western blot结果显示加药组细胞中与增殖有关蛋白PCNA的表达量显著下调,与凋亡有关蛋白Caspase-3、Caspase-8、Caspase-9、PARP被剪切激活。结论:柯里拉京对宫颈癌Hela细胞有明显的抑制作用,其抑制作用与柯里拉京诱导细胞周期阻滞及凋亡有关。
To study the effect of corilagin on proliferation and apoptosis of human cervical cancer Hela cells. After Hela cells were treated with different concentration of corilagin,the inhibitory effect of corilagin on Hela cells was detected by MTT assay and Colony formation assay. Morphological changes were observed by inverted microscope. The cell cycle and mortality were analyzed by flow cytometry. The expressions of PCNA,Caspase-3,Caspase-8,Caspase-9,PARP proteins were examined by Western blot. Corilagin significantly inhibited the proliferation of cervical cancer Hela cells in a dose-dependent manner; the result of flow cytometry indicated that corilagin could induce Hela cells to undergo S stage cycle arrest; Western blot showed that the expression of proliferation-associated PCNA protein was remarkedly downregulated and apoptosis-associated Caspase-3,Caspase-8,Caspase-9,PARP proteins were sheared and activated in Hela cells. It shows that corilagin has a significant inhibitory effect on cervical cancer Hela cells,and its inhibitory effect is associated with cell cycle arrest and apoptosis.
