膀胱癌组织自体荧光特征分析及其机制研究

摘要 目的:分析膀胱癌组织自体荧光光谱特征及其与正常组织的差异;探讨大鼠膀胱正常组织和癌组织中Ⅳ胶原和组织蛋白酶B的表达差异和相关性。方法:采集了47例膀胱癌组织和47例正常组织的自体荧光光谱,分析其荧光光谱差异及其特征。用免疫组化染色的方法检测正常组、膀胱癌组各25例组织标本中Ⅳ胶原和组织蛋白酶B的表达,比较各组间差异,并进行相关性分析,同时检查组织蛋白酶B mRNA的表达状况。结果:94标本共采集了300~800nm波长范围的癌组织和正常组织自体荧光光谱,发现正常组织的荧光强度在任意波长下均高于癌组织,正常组织的峰强度明显高于癌组织(P<0.05)。免疫组化染色显示正常组、癌组中的基底膜均有Ⅳ胶原表达,但膀胱癌组织中表达减少,与正常膀胱组织组比较,差异有显著性(P<0.05)。组织蛋白酶B在膀胱癌组织中表达较正常膀胱组织表达增加,差异有显著性(P<0.05)。结论:膀胱正常组织、癌组织的自体荧光光谱特征存在差异;在膀胱正常组织及癌组织中,大鼠膀胱癌组织中Ⅳ胶原表达减少;大鼠膀胱癌组织中组织蛋白酶B表达增加,可能导致两组组织的自体荧光差异的原因之一。 Objective:To analyze the characteristics and difference of the laser-induced autofluorescence spectra of normal bladder tissue and bladder carcinoma tissue and find a method for discrimination among normal bladder tissue and bladder carcinoma tissue.To investigate the difference in collagenⅣande cathepsin B among normal bladder tissueand carcinoma tissue and the correlation between collagenⅣ ande cathepsin B expression in bladder carcinoma tissue of rats.Methods:Adopting laser-induced autofluorescence detection analytical system to collect 47 autofluorescence spectra of cutting normal bladder tissue and bladder carcinoma tissue in surgery in 300-800 nm.25 bladder specimens of rats were involved to detecte collagenⅣande cathepsin B expression in eaeh group:normal bladder tissue and bladder carcinoma tissue from the ratcolorectal carcinoma model.The collagen Ⅳ and cathepsin B was detected with imrnumohistochemical staining.The correlation of the results was studied.Cathepsin B RNA expression was detected with reverse transcription-Polymerase chainreaction(R-TPCR).Results:Of94 specimens detected by laser-induced autofluorescence,47 were normal and 47 carcinoma tissue.All normal tissue fluorescence intensity was high than carcinoma tissue on the same wavelength.There was a char wave crest at 452 nm respectively.Normal tissue of peak intensity was obviously high than carcinoma tissue(P〈0.05).CollagenⅣexpression existed in all specimens in different degrees,it decreased carcinoma group,and there wass significant differenece compared with the normal group and hyperplasia group.The expression of cathepsin B in carcinoma group increased compared with normal group(P〈0.05).Conclusions:Normal tissue and carcinoma tissue of autofluorescence intensity were difference,laser-induced bladder tissue autofluorescence spectra may be useed in distinguishing Normal tissue and carcinoma tissue.The expression of Collagen Ⅳ decreased in bladder carcinoma compared with the normal bladder tissue.The expression of cathepsin B increased in bladder carcinoma compared with the normal bladder tissue,Possible induce difference of one of reason for autofluorescence in normal bladder tissue and bladder carcinoma tissue.
