摘要
目的研究两种艾灸血清对实验性类风湿性关节炎(rheumatoid arthritis,RA)大鼠滑膜成纤维细胞体外增殖及滑膜成纤维细胞肿瘤坏死因子受体相关因子6(TRAF6)表达的影响,探究艾灸血清对实验性RA大鼠抗炎效应的作用机制。方法将36只SD大鼠随机分为3组,即模型组、直接灸组、隔姜灸组,每组12只,每组大鼠制备RA模型。造模成功后,模型组不予以任何治疗,直接灸组、隔姜灸组分别给予直接灸法、隔姜灸法刺激RA模型大鼠的"肾俞"、"足三里"。疗程结束后,取腹主动脉血并离心取血清,用含有各组血清培养基培养RA模型滑膜成纤维细胞48 h,用Ed U试剂盒检测各组血清培养的滑膜成纤维细胞增殖情况并用Western blot检测培养48 h时各组细胞TRAF6的表达量。结果艾灸血清培养的RA滑膜成纤维细胞的增殖与模型组血清培养的RA滑膜成纤维细胞相比,细胞体外增殖受到抑制,细胞TRAF6表达量明显下降,以隔姜灸效果更为显著。结论艾灸血清可能通过抑制细胞TRAF6表达从而抑制RA滑膜成纤维细胞的增殖,发挥其抗炎作用。
Objective To explore the anti-inflammatory effect of moxibustion serum on experimental RA rats by investigation of its influence on synovial fibroblast proliferation and TRAF6 expression in vitro. Methods Thirty-six SD rats were randomly divided into three groups including the model group,direct moxibustion group and ginger moxibustion group. After RA rat models were successfully established,the model group did not receive any treatment. The direct moxibustion group and ginger moxibustion group were stimulated with the "Shenshu" and "Zusanli",respectively. After treatment,abdominal aorta blood was taken and serum was collected by centrifugation. RA model synovial fibroblasts were isolated and cultured with each group of serum media for 48 h. The proliferation of synovial fibroblasts was detected by Ed U kit,and the expression of TRAF6 was detected by western blot. Results Compared with the model group serum,the proliferation of synovial fibroblasts and the expression of TRAF6 were significantly decreased in two moxibustion serum groups,especially in ginger moxibustion serum group. Conclusion Moxibustion serum may exert an antiinflammatory effect through inhibiting the TRAF6 expression and synovial fibroblast proliferation.
