摘要
玉米单倍体和DH系的研究以及应用越来越被重视,但单倍体加倍效率低致使其利用价值在一定程度上受到限制,该研究通过对比不同方法(自然加倍、浸种法、浸芽法、注射法)不同处理对玉米单倍体的加倍效果,以期探索出较好的单倍体加倍方法。结果表明:自然加倍率较低,仅为2.5%;相比浸芽法、注射法,浸种法加倍效果尤为突出,且0.2%浓度的秋水仙素,浸种处理12 h效果最佳,其植株成活率、散粉率、结实率分别达到了78.57%,57.14%,19.64%。
The research and application of maize haploid and DH lines have been paid more and more attention. However,the low effect of haploid doubling makes the utilization value of maize haploid be limited to a certain extent. In this study,to explore better haploid doubling methods, these haploids were dealed with different treatments and different methods(natural doubling method, soaking seed method, soaking bud method and injection method). The results showed that the natural doubling rate was relatively low, only 2.5%;compared with natural doubling method, soaking bud method and injection method, soaking seed method was especially effective. And soaking seed 12 h with 0.2% concentration of colchicines had the highest doubling efficiency, and its survival rate, loose powder rate and seed setting rate reached 78.57%, 57.14% and 19.64% respectively.
