摘要
以杉木(Cunninghamia lanceolata)嫩茎为实验材料,开展了组织培养研究。结果表明,杉木嫩茎用0.1%的HgCl_2试剂消毒9 min效果最好,污染率和死亡率低,成活率达到80%。最佳增殖培养基为MS+6-BA1.5 mg/L+NAA0.3 mg/L+3%蔗糖,增殖系数达到4.2。最适宜杉木组培苗生根的培养基为1/2MS+NAA0.1 mg/L+IBA1.5 mg/L+2%蔗糖,生根率为80%。移栽30 d后,成活率高达88%。
The tender stem segments of Cunninghamia lanceolata was used for tissue culture and micropropagation. The results showed that 0.1% mercuric chloride solution(9 min) was appropriate, the pollution and mortality rate was very low,the survival rate was 80%.And the best medium for subculture was MS+6-BA1.5 mg/L+NAA0.3 mg/L+3%S, the value-added coefficient reaches 4.2.The most suitable medium for rooting was 1/2 MS+NAA0.1 mg/L+IBA1.5 mg/L+2%S,the rooting rate was 80%.After 30 days, the survival rate was more than 88%.
