1株产类胡萝卜素盐盒菌的分离鉴定及培养条件优化

摘要 对分离自运城盐湖黑泥中的一株产类胡萝卜素嗜盐菌G14进行分离鉴定,经菌落形态观察、生理生化特性及16S rRNA基因序列分析,确定该菌株隶属于盐盒菌属(Haloarcula sp.)。通过单因素与正交试验对菌株G14的培养条件进行优化。结果表明,菌株G14最佳培养条件为NaCl质量浓度0.23 g/mL,初始pH值为7.0,43℃光照培养。在此优化条件下,培养的菌液OD_(600 nm)值达2.24,类胡萝卜素产量为0.65 mg/L。 A halophilic strain G14 with carotenoid yield that isolated from the black clay of Yuncheng Salt Lake was identified through colonial morphologies, biochemical reactions and 16 S rRNA sequence analysis, and results showed that the strain was Haloarcula sp. The culture conditions of strain G14 were optimized by single factor and orthogonal experiments. The results showed that the optimal culture conditions of strain G14 were NaCl mass concentration 0.23 g/ml, initial pH value 7.0 and light culture at 43 ℃. Under the conditions, the maximal OD(600 nm)of the bacteria solution reached 2.24, and carotenoid yield was 0.65 mg/L.
