摘要
目的雷帕霉素是临床上治疗肿瘤的一线药物,但其在神经内分泌肿瘤中的治疗机制尚不清楚。本文主要探讨雷帕霉素能否调节Caspase-9和Ki-67表达水平,从而发挥抗神经内分泌肿瘤的作用。方法 488例神经内分泌肿瘤患者随机分为对照组和雷帕霉素干预组。免疫组化和western blot检测Caspase-9和Ki-67的表达情况。结果免疫组化结果表明,Caspase-9和Ki-67之间具有明显的相关性(P<0.05)。经雷帕霉素治疗后,神经内分泌肿瘤患者的Caspase-9的表达水平显著上升,Ki-67的表达水平显著下降,与对照组相比,差异具有统计学意义(P<0.05)。结论雷帕霉素可能通过调节Caspase-9和Ki-67的表达水平而发挥其抗肿瘤作用。
ObjectiveRapamycin is the first-line drug for the treatment of tumors in clinic,but its therapeutic mechanism in neuroendocrine tumors is still unclear.This article mainly discusses whether rapamycin can regulate the expression level of Caspase-9 and Ki-67,thus exerting the effect of anti-neuroendocrine tumors.Methods 488 patients with neuroendocrine tumors were randomly divided into control group and rapamycin intervention group.Immunohistochemistry and western blot were used to detect the expression of Caspase-9 and Ki-67.Results The results of immunohistochemistry showed that there was a significant correlation between Caspase-9 and Ki-67(P〈0.05).After treatment with rapamycin,the expression of Caspase-9 in patients with neuroendocrine tumors increased significantly,and the expression level of Ki-67 decreased significantly.Compared with the control group,the difference was statistically significant(P〈0.05).Conclusion Rapamycin may exert its anti-tumor effect through regulating the expression of Caspase-9 and Ki-67.
